The pigment of edulis is an anthocyanin pigment, it is mainly existed in the stems, leaves and fruit of the edulis. In recent years, with the understand of the natural food pigment and the constant pursuit of quality of life, people realize that edulis pigment with the high nutritional and medicinal value, so the development and application of edulis pigment are concerned by more and more people.This paper is mainly research of the pigment of edulis, study on the extraction, purification and stability of the pigment of edulis.This paper adopts four different methods to extract pigment from edulis. The optimal method and optimal condition are ascertained by comparing the content of the pigment.Single factor test and orthogonal experimental design method are used to determine the optimum condition of alcohol extraction, the optimum conditions are:30℃,50% ethanol, liquid ratio 1:10, time 5 h, under these conditions the extraction rate is 85.31%; the optimum conditions of enzymatic extraction are:pectinase dosage 200μL/L, reaction temperature 45℃, hydrolysis time 3.5 h, pH=5, the extraction rate under the conditions is 92.24%; the optimum conditions of ultrasonic extraction are:20℃, liquid ratio 1:20, power 200 W,50 min, extraction rate is 90.17%; ultimately determine the best method of extraction is enzyme-ultrasonic, extraction rate is 94.86%.Purification of extracts selects the macroporous resin purification, determine the AB-8 macroporous resin is the best adsorbent by resin adsorption, mainly studies the effects of sample solution pH, eluent amount, eluent space velocity, eluent concentration. Determine the optimum condition are:the sample solution pH=3, the 60% ethanol, eluent amount 2 BV, eluent space velocity 1 SV, the refined purity of red pigment edulis up to more than 50%.Use the edulis red pigment for the stability test, the results show that the red pigment is relatively stable under the acidic conditions; light has the great effect of the red pigment; the properties of pigment is stable under 60℃,60℃or more the pigment molecules vulnerable to decomposition; Ca2+,Zn2+,Mg2+have the little effect for the red pigment, Cu+,Al3+ have the effect of the color solid and protection, Fe3+ has the great effect of the pigment, easily to make the pigment decomposition.Through the IR detection, HPLC detection testing methods to determine the major components contained in the pigment is cyaniding-3-glucoside.In this study:using enzyme-ultrasonic extraction of edulis red pigment and macroporous adsorption resin for purification of red pigment, the effect is obviously, suitable for industrial production, has the great significance of the natural plant pigment.
