Pinus tabulaeformis polllen was called "Song Huang''in ancient times,it was the sperm cell of pinus tabulaeformis,its composition was balanced and it was a medicine combination of pure natural food materials.pinus tabulaeformis can anti-aging,anti-fatigue,reduce blood fat and strengthen body immunity,it has high value to exploit and utilize.The pinus tabulaeformis pollen polyphenols were extracted by ultrasonic with ethanol,based on the results of single factor experiment,the significant impact factors were selected through the Plackett-Burman,the polyphenol content was as the response target,the selected factors were optimized by the response surface.The positive comparison was Vc,the reducing power of pinus tabulaeformis pollen polyphenols and their scavenging effect on DPPH·,·OH,O2-·were researched by Fe3+-reducing method,DPPH· reducing method,salicylic acid colorimetry and pyrogallol autoxidation method,antioxidant activities of polyphenols from pinus pollen were evaluated synthetically,the experimental results were as following:(1)The optimal extraction conditions of pinus tabulaeformis pollen polyphenols:the ethanol volume fraction,extraction temperature,liquid ratio and extraction time were used to do the single factor experiment,based on the results of single factor experiment,through the Plackett-Burman selection,extraction temperature,liquid ratio and extraction time were determined are the significant impact factors,the response surface analysis was chosen to optimize the extraction process of pinus tabulaeformis pollen polyphenols,it is concluded that the optimal extraction conditions were:the ethanol volume fraction 80%,liquid ratio 1:32 g/mL,extraction time 31 minutes and extraction temperature 51 ?.Under the conditions,the yield of total phenols from pinus tabulaeformis pollen can be up to 6.735 mg/g.(2)The separation and purification of pinus tabulaeformis pollen polyphenols:after rotary evaporated and enriched,the crude extract of pinus tabulaeformis pollen polyphenols were separated and purified by the preprocessed AB-8 macroporous adsorption resin,the water-dissolved crude extract of pinus tabulaeformis pollen polyphenols were vibrated and adsorbed by the weighed AB-8 macroporous adsorption resin,then the resin was desorbed by adding ethanol,the pinus tabulaeformis pollen polyphenols were stored as backup after rotary evaporated and enriched without no ethanol flavor.(3)The antioxidant activity of pinus pollen polyphenols:the positive comparison was Vc,the reducing power of pinus tabulaeforrmis pollen polyphenols and their scavenging eff-ect on DPPH·,·OH,O2-·were researched,the reducing power,DPPH·clearance,·OH clearance and O2-·clearance were the antioxidant indicators.In the range of experimental concentration,the pinus pollen polyphenols have stronger antioxidant activity,the reducing power is the strongest,the scavenging effect on DPPH·and·OH are weaker,the 02-·'s is the weakest,that is reducing power>DPPH·clearance>·OH clearance>O2-·clearance,but all antioxidant indicators are better than Vc.
