Flavonoidshaveeffects of anti-cancerand anti-oxidation,and so on.theyhave great application value in fields of food and medicine.usingwaste of Huyou peelsas raw materials duringHuyou processing infood plant,This paper studied methods of extraction,isolation,determination offlavonoidsinHuyoupeels.Finally,the insoluble dietary fiberwas extracted from residue of pomelo peels,and alkali treatment process was discussed.This paper researched the influencesof extraction process conditions of ethanol on the extraction ratio of flavonoids,the best extraction conditions were obtained:ethanol volume fraction was 70%,extraction time was 100 minutes,extraction temperature was 60?,ratio ofmaterial to solvent was 1:10(g/ml).Under the optimum process conditions as mentioned above,the flavonoids were extracted,under the conditions of 2-5?and crystallization for 48 hours,the crystal was obtained from extracting solution after being concentrated and through the way of washing by distilled water and recrystallization,the purity 70.36%flavonoids compound were obtained,the yield was 1.13mg/g.by alkali extraction and acidprecipitation method,the flavonoids with purity of 34.23% were obtained.The method of isolating flavonoids was studied by silica gelcolumn chromatography.Firstly,the flavonoids were extracted through ethanol impregnationmethod.extracting solution was analyzed through thin layer chromatography,using dichloromethane–methanol(4:1)as developing solvent and Iodine steamas color agent,6 color spots and 1 ribbon were separated,this solvent system was determined as eluent for silica gel column chromatography.Using dichloromethane-methanol(10:1,6:1,3:1,0:1),Flavonoids were isolated from impregnation liquid in the way of gradient elution.Identified by the means of uv-spectrophotometry,mass spectrometry,thin layer chromatography(TLC),High Performance Liquid Chromatography(HPLC),the flavonoids were determined as a mixture of naringinand neohesperidin,The flavonoids purity was 82.47%,the yield was 3.12mg/g.in this method recovery rate of ethanol was investigated,total recovery reached 74.1%.Taking pomelo peel slag generated after extraction of flavone as raw material,this study investigated the effect of alkali treatment process conditions on the extraction of insoluble dietary fiber from residue.The optimum condition was found: the ratio of material to sodium hydroxide solution is 1:9(g/ml),the concentration of sodium hydroxide was 0.5mol/L,the extraction time was 1.5 hours,the temperature was 50?,under this condition the yield was 60%,Swelling powerwas 6(ml/g),water holding capacity was 4.44(g/g).
