In China,the annual yield of wheat bran is considerable,which can provide large amounts of dietary fiber,and production technology of soluble dietary fiber(SDF)and oligosaccharide from wheat bran has been developed.Compositions obtained by different techniques will be different,therefore,physiological functions vary as well.Two methods,enzyme-alkali extraction technique and solid state fermentation technique,were adopted to extract SDF.Bran oligosaccharide was extracted by extrusion-composite enzyme technique and alkali extraction-composite enzyme technique.The result was that the yield of SDF through alkali extraction could be reached 5.4%and the content of SDF was 78.76%,and the yield of SDF through solid-state fermentation could be reached 11.1%and the content of SDF was 83.93%.In addition,extrusion ?composite enzymatic preparation of oligosaccharide was at a rate of 28.33%,and the alkali-composite enzymatic technique was 21.63%.The molecular weight distribution of the four extracts was determined by HPLC which proved two kinds of soluble dietary fiber does not contain low poly SDF.The content of poly SDF by the solid state fermentation was 2.41%,while the content of poly SDF by composite enzyme-alkali technique was 27.74%.The molecular weight of oligosaccharide by extrusion-composite enzyme was 940D,and the content is about 62%.The molecular weight of alkali-composite enzyme oligosaccharide was 1080D,and the content is about 57%.SEM showed that the structure of four extracts was reticulate structure and flake,In particular,extrusion technology made the surface of oligosaccharide smoother.The laxative function of four extracts was compared in vivo tests,we found that composite enzyme-alkali SDF(ig.5g/k · bw)and extrusion-composite enzyme oligosaccharide(ig.0.8g/kg · bw)could significantly shorten the time of the first black dejection and increase the weight of defecation.In terms of intestinal bacteria group,solidstate fermentation SDF(ig.5g/kg · bw)and extrusion-composite enzyme oligosaccharide(ig.0.8g/kg · bw)could improve the number of intestinal beneficial bacteria---Lactobacillus.After using single factors experiment and orthogonal experiment,in combination with sensory evaluation,we got the optimal formula for making oligosaccharide jelly drops.The optimal formula was:sugar 55%,carrageenan 2.5%,oligosaccharide 4%and citric acid 0.6%.
