Lignan is produced by the dimerization of cinnamic alcohol dimer reaction of macromolecular compounds,and it's basic structure of phenylalanine.Lignan is a polyphenolic substance extracted from flax,which is the largest plant in the known plants.The study found that lignans can be combined with a variety of enzymes to inhibit the synthesis of human ovarian hormone,through this inhibition can reduce the risk of women suffering from breast cancer and other gynecological diseases.In addition,lignans attract more and more scientists's attention because of its anti-cancer,anti-fungal and the removal of free radicals and many other functions.In this study,lignans are the objectligne;Cottonseed hull was repleced by flaxseed to cultivate F.velutipes.Flammulina species will break down linseed shellsin the growth phase.We can regulate the flax shell and other raw materials of different materials than the normal growth of F.velutipes,not only the growth of F.velutipes was in good condition,but also make the linseed shell decomposition of the utilization rate of the maximum..The lignan substances in the flaxseed substrate were separated?identified and quantitative analysis by RP-HPLC/UV and UPLC/MS/MS,respectively.At the same time,antioxidant,hypoglycemic and antifungal activity of lignans were determined by DPPH antioxidant,a-glucosidase inhibitor and Oxford cup method respectively.In order to providing basic biological data for further study of the effect of lignan in vivo activity experiments or animal model experiments.Finally,the biodegradation process of F.velutipes strains to lignin was studied in different time periods,and it was expected that the trend of lignin contents could be used as a new extrapolation criterion of lignin biodegradation.In the first part of this paper,flaxseed is used to cultivate F.velutipes instead of cottonseed hull and sawdust.By adjusting the ratio of different materials,the optimum proportion of material ratio is flaxseedl5%,Bran 5%%,Glucose 5%,KH2P04 0.1%,MgS04 0.05%,CaC03 0.2%and water 74.65%.Linans were extracted by 80%methanol,and identified by HPLC/UPLC-MS.Seven different lignan monomeric products were obtained,which were as follows:secoisolariciresinoldi-glucoside(SDG),(b)secoisolariciresinol(SECO),(c)matairesinol(MATA),(d)pinoresinol(PI NO),(e)Pinoresinol diglucoside,(f)ferulic acid glucoside(FeAG),(g)p-Coumaric acid glucoside(CouAG).In the secondary spectrum of UPLC/MS/MS,the secondary spectra of the seven substances are very clear.The contents of seven substances were determined by relative quantification method:FeAG 16.521mg/g?PINO 16.485mg/g?SECO 8.387mg/g?MATA 15.930mg/g?CouAG6.21mg/g?SDG12.874mg/g?Pinoresinol diglucoside 5.101mg/g,respectively.Meanwhile,the total phenolic content of lignans in the flaxseed matrix was 617.5mgGAE/g.In the second part,the antioxidant,hypoglycemic and antifungal activities of lignans were determined by DPPH method,a-glucosidase inhibitor method and Oxford cup method,respectively.The bioactivities of lignans were studied in vitro using a-glucosidase,DPPH oxidation and Oxford cup method.The results showed that lignans had good hypoglycemic,antioxidation and the bacteriostasis of Escherichia coli.In this study,the IC50 of lignas was 3.41±0.31?g/mL and 4.14±0.55?g/mL,respectively.In the third part of this paper,the main purpose of this study was to provide suitable carbon and nitrogen sources for the growth of F.velutipes,as well as good environmental conditions,which was beneficial to the normal growth of F.velutipes and release acid-base products.The active components of lignans can be released from lignin,and the dissociation of the whole structure of flaxseed cell wall and the dissociation of the whole structure of flaxseed cell wall.And the lignans macromolecule polymer was depolymerized to form lignan-like compounds.At the same time,the contents of various substances in the linen shell matrix were determined and analyzed by DPPH antioxidant method and a-glucosidase inhibition.The results showed that lignans could improve the biodegradation of lignin through the biodegradation of lignans.The results showed that lignans could be used as biodegradable microorganisms.The change of content in the process can be used as the standard to extrapolate the degree of biodegradation of lignin,which can provide basic biological parameters for further study on the pathways and mechanism of F.velutipes in biodegradation of linseed and other lignin.
