Development Of The Colloidal Gold Immunochromatographic Rapid Test Strip For The Detection Of Deltamethrin

Pyrethroids pesticides are an important class of broad-spectrum insecticides,but recent studies have shown that long-term low dose exposure of pyrethroids pesticides can cause chronic disease such as immune system toxicity,genetic toxicity and cardiovascular toxicity.Therefore,the effective supervision of pesticide residue in agricultural products has become a widely concern.Immunochromatographic(ICG)test strip are becoming increasingly common as detection method due to their sensitivity,simplicity,lower cost and shorter time,no need of technical skill to interpret the result and overall it is more affordable than other conventional methods.The immunochromatographic method is used to give visual color indication of the presence or absence of the analyte.However this technology are not easy reproducible by other users due to the complicated procedures in construction.In this study a rapid,simple and sensitive immunochromatographic test strip has been developed for testing residues of deltamethrin in river water sample.The experiment using caprylic acid and ammonium sulphate to purify the monoclonal antibody against deltamethrin.30 nm colloidal gold was prepared by sodium citrate method,the optimal pH value of colloidal gold solution and the amount of antibody were 8 and 30?g/ml.Nitrocellulose(NC)membrane was used as carrier through optimization of colloidal gold immunochromatographic strip test the best working condition:coating antigen was diluted to 1:20,Goat anti-mouse secondary antibody with optimal dilution of 1:50,nitrocellulose membrane 135s coated with PBS solution,the best standard dilution was PBST solution containing 5%methanol,gold standard pad was blocked with 0.01%BSA 0.05%SDS-L of PBS buffer solution.The visual detection limit of deltamethrin colloidal gold immunochromatographic assay was 10ng/ml,the detection limit is less than 10min.the detection limit of river water sample was 200ng/ml.The process of detection is simple,rapid and convenient the results can be visually read by eyes without any instrumentation,the detection method is simple to operate high sensitive and can be used in the field as screening test for the detection of deltamethrin residue.
