Walnut resources are rich in our country,diaphragma of juglans regia in the walnut are often discarded in the process of walnut processing,it has not been fully utilized,its necessary to research and develop diaphragma of juglans regias medicinal value and health care value.To use the resources fully,this article regard diaphragma of juglans regia as raw material,extracted flavonoids compounds and study the best extraction technology,and its antioxidant activity were studied,to provide theoretical basis for the development and utilization of diaphragma of juglans regia.By ultrasound-assisted heat extraction,single factor test and response surface optimization experiments were studied to determine the optimum extraction:ethanol concentration was 50%,extraction temperature was 50?,solid-liquid ratio was 1:40,time was 40min.By the optimized extraction process,the extractions of total flavonoids from diaphragma of juglans regia are 115.54 mg/g.Through the static adsorption experiments and analytical experiments,from four kinds of macroporous resin,we selected AB-8 resin to separate and purify flavonoids from diaphragma of juglans regia.Then,50%ethanol was used as eluent to elute resin dynamically.Affording elution profile,and ultimately determining flavonoids purity in extraction increased from 41.78 percent to 61.34 percent.Using Fourier Transform Infrared Spectroscopy walnut extract diaphragm,we detects the characteristic absorption of flavonoids.By liquid chromatography-mass spectrometry technique,results showed that the extract contains luteolin and quercetin.Ant-oxidant studies in vitro suggest that flavonoids from diaphragma of juglans regia have reducing power,hydroxyl radical scavenging,DPPH · clearance and total antioxidant capacity,at the same concentration,flavonoids extractions from diaphragma of juglans regia antioxidant capacity is lower than Vc,and greater than BHT.Use D-galactose induced aging rat model,oraling flavonoids from diaphragma of juglans regia and vitamin E eight weeks,the water maze experiments was carried out,and conduction showed that the model was successful,each dose of the test group of mice space exploration capability,memory capacity are significantly stronger than the model group,and the middle dose group was the best.Using this model to study the effect of different dose groups' flavonoids extraction from diaphragma of juglans regia on antioxidant system in mice.The results showed that:flavonoids from diaphragma of juglans regia can significantly increase the total number of the total number of leukocytes and lymphocytes.Walnut diaphragm flavonoids extract significantly or very significantly increased serum,liver and brain tissue of mice SOD,T-AOC and GSH-Px enzyme vitality,reduce the content of MDA.Dose(100 mg/kg),high-dose(200 mg/kg)and VE(50 mg/kg)were better.The relative content of fatty acids in mouse brain were measured,D-galactose can reduce the content of unsaturated fatty acids in the brain,while the low dose group and high dose group compared with the model group can weaken the role of the D-galactose,by improving not saturated fatty acids,in particular EPA and DHA fatty acids,play a role in delaying aging mice.Through the brain tissue slices,flavonoids from diaphragma of juglans regia have a protective effect on hippocampal tissue membrane.
