Dyslipidemia induce myocardial infarction,coronary heart disease,stroke and other cardiovascular and cerebrovascular diseases risks.It is necessary to develop a lipid-lowering soft capsule of prevention and mitigation dyslipidemia in such circumstances.This study was researched on its own separate cholesterol lowering effectively Lactobacillus plantarum M1-UVs29 for starting strain,Lactobacillus plantarum M1-UVs29 and rice bran oil dual health care function of lowering cholesterol is researched.Research is mainly aimed at product lipid-lowering soft capsule and explored the effect of lipid-lowering by in vitro and in vivo tests.The main research contents are as follows:(1)Lactobacillus plantarum M1-UVs29 freeze-drying protective agent recipe was selected and optimized by Plackett-Burman,the steepest ascent and Box-Benhnken design in this study.The best ratio of protective agent is: skim milk 13.00%(w/v),trehalose 7.60%(w/v),sorbitol2.70%(w/v),the bacteria powder survival rate was 86.28%,add protectant bacteria powder than without protectant bacteria powder in artificial gastric juice,artificial intestinal juice,storage capacity and heat treatment show a significant protection(P < 0.05).Lactobacillus plantarum M1-UVs29 powder added protectant provides some references for functional food.(2)Lactobacillus plantarum M1-UVs29 added protective agent recipe and rice bran oil in water enzyme method were made of soft capsule.According to the production process requirement,lipid-lowering best ratio of soft capsule contents as follows: Lactobacillus plantarum bacteria powder 14.00%,oligofructose 14.00%,rice bran oil 69.97%,wax content2.00% and antioxidant content 0.03%.Soft capsule sensory,physical and chemical indicators,hygienic index,accelerated test and stability test accord with quality standards.Lipid-lowering soft capsule was qualified.(3)Lipid-lowering soft capsule content perform the experiment in vitro.Content of lipid-lowering soft capsule lower cholesterol,and fat adsorption capacity experiment results in vitro show that lower cholesterol capacity of more than 59.23 % and fat adsorption capacity can reach 21.14% in vitro;experiment indicated that the contents can have the effect of lipid-lowering and cholesterol-lowering in vitro.(4)To lipid-lowering soft capsule content as raw materials,the animal experiment wascarried out on the C57BL/6J mice.And indexes of weight,mesenteric fat weight ratio,epididymal fat weight ratio,kidney weight ratio,liver weight ratio,intestinal flora,serum TC,TG,LDL,HDL,serum INS,TNF-?,THARS,TG in the liver,FAS,CPT-1 of liver,liver pathological was determinated and pathological was analysised.The soft capsule dosage group compared with high-fat group,analysis of variance was significantly(P < 0.05),animal testing showed that the soft capsules play a positive role on cholesterol-lowering.Through the above test results were showed that the lipid-lowering soft capsule formulations were reasonable,Lactobacillus plantarum M1-UVs29 active bacteria number accord with national quality standard.Stability is good,and it has effect on prevent and alleviate dyslipidemia.
