A novel method for sample preparation was used for the quantitative analysis of levofloxacin and vitamin B1 in pharmaceutical preparations by diffuse reflectance Fourier transform infrared spectroscopy?DRIFTS?technique and satisfying results were obtained.The method of DRIFTS,one kind of infrared analysis method,was characterized by directly measuring powder samples and no special sample pretreatment.It was also applied for the samples with no adequate transparency or surface smoothness.Samples would not be damaged and polluted during the process of analysis.The method of DRIFTS could be utilized for qualitative analysis as well as quantitative analysis and the simultaneous analysis and test of the multicomponent in compound could also be achieved.Ofloxacin is a third-generation quinolone antibacterial medicine synthesized chemically.It's broad-spectrum,highly active and mainly available for the low and moderate infections of respiratory,digestive,urinary and other systems caused by various sensitive bacteria.Irrational and high-dose use of ofloxacin may lead to drug resistance or multiple toxic side-effects,therefore the quality control of ofloxacin pharmaceutical preparations is significant.The stretching vibration absorption peak of ketone carbonyl at 1620cm-1 was selected as the quantitative analysis peak during determining the content of levofloxacin in preparations by DRIFTS.The peak area in the region 15801655cm-1 showed a good linear relationship with the mass fraction of levofloxacin in the range of 0.20%1.20%.The correlation coefficient of the standard working curve was 0.984 and the relative standard deviation?RSD?of the determination results of the samples was ?5.6%.The interference of insoluble starch in the auxiliary material to analytic determination could be absolutely eliminated by filtering operation in the process of the novel sample preparation.The interference of less soluble starch to analytic determination could be eliminated through the application of the standard working curve of the levofloxacin standard sample containing soluble starch.The recoveries were 97.4%100.3%.Vitamin B1 is an essential nutrient element for human body,mainly used for the prevention and treatment of beriberi,neuritis,malnutrition etc..It can not be synthesized and stored in the human body,only supplemented from food and drug intake.Accurate determination of the content of vitamin B1 in food and medicine is of great significance to healthy diet and medical care and treatment.The content of vitamin B1 in tablets was determined by DRIFTS in this paper.Theabsorption peak at 1659cm-1 was chosen as the quantitative analysis peak.The peak area of region16301690cm-1was used to evaluate the absorbance of samples not containing tartaric acid and citric acid in tablets accessories.And the single wavenumber at 1654cm-1 was used to evaluate the absorbance of samples containing tartaric acid or citric acid.The mass fraction of vitamin B1 showed a good linear relationship with the absorbance,the correlation coefficients were 0.982 and0.984 respectively and the relative standard deviation ? 6.2%.The interference of soluble impurities could be excluded by choosing the appropriate absorbance evaluation method and the baseline correction range and thus the accurately quantitative analysis of vitamin B1 in pharmaceutical preparations could be achieved.The recoveries were 97.2%100.6%.The results of three kinds of commercial available levofloxacin preparations and four kinds of commercial available vitamin B1 tablets obtained by DRIFTS are accurate and in accordance with those obtained by ultraviolet spectrophotometry?UV?.The method of DRIFTS applied for quantitative analysis is simple,rapid,accurate and reliable.It is characterized by simple sample pretreatment,less dosage of reagents and specimens,strong analysis characteristic and no waste.In addition,the method of DRIFTS has low analysis expenses and wide application.Overall,the method of DRIFTS is one kind of environmental friendly analysis method.The sample and KBr can be mixed more evenly,the specular reflection light can be effectively eliminated and the sensitivity and precision of the analytical results will be improved by using the novel method for sample preparation;the samples can be measured in a lower concentration range and thus the linearity of the standard working curve is so much the better;the accurate determination of the multi dosage forms can be realized and the scope of application can be expanded.
