OBJECTIVE study the determination of the main chemical components in the purple yam,the best extraction method of anthocyanin,the stability of anthocyanin and the screening of anti-tumor activity.Methods ? The protein content was determined by Coomassie brilliant blue method,and the content of total sugar was determined by water extraction-alcohol precipitation method.? Purple yam washed,sliced,the use of acidic ethanol extraction method,ultrasonic assisted extraction method,heating assisted extraction method for crude extraction.By measuring the absorbance at 530 nm,compared to take the maximum absorbance value of the method used to extract.And then the single factor experiment and orthogonal experiment were carried out to determine the optimal conditions for extracting anthocyanin.? Anti-tumor activity of HepG2 hepatoma cells,MDA-MB-231 breast cancer cells and RAW264.7 macrophages were detected by cck8 method.Results ? The protein content was 2.0%and the polysaccharide content was 28%.The optimum extraction parameters of anthocyanins in purple yam were:60 ?,2%HCl-40%ethanol,extraction time 40 min,ratio of material to liquid 1:10.? The IC50 of anthocyanin to HepG2 hepatoma cells was 223?g/mL,the IC50 of breast cancer cells was 112.56?g/mL,and the IC50 of RAW264.7 macrophages was 26.32?g/mL.ConClusion ? Compared with the common yam varieties,the polysaccharide content of Ziyan medicine was higher.? the use of acidic ethanol solution can be effective extraction of anthocyanins in purple yam.At the same time,the most experimental factors affecting the extraction of anthocyanins were the ratio of material to liquid.Under acidic conditions,the stability of the pigment is high and should be extracted under acidic conditions.Should control the temperature of anthocyanin extraction.Place the pigment extract in a brown bottle for storage,as it is more sensitive to light.The inhibitory effect of anthocyanin on HepG2 hepatoma cells,MDA-MB-231 breast cancer cells and RAW264.7 macrophages was different,especially in RAW264.7 macrophages.
