Vitamin A?D?E were three kinds of fat soluble Vitamins,they were essential nutrients that play important role in maintaining a body worked healthily and guaranteeing the normal reproduction function.The primary source of FSVs is the food we take in our daily life.It's do important to evaluate the content of FSVs in food.In this work,a new method was developed to simultaneously determine all 8 vitamin E and Vitamin A in food.Vitamin E isomers and Vitamin A were separated by BEH Amide column(150 mm×3.0 mm,1.7 ?m)with [n-hexane:(methyl-tert-butyl ether,tetrahydrofuran,methanol,20:10:1,v/v/v),90:10,v/v)] as mobile phase.Ultraviolet detector and fluorescence detector were employed to detect the target compounds.The method is verified can rapid baseline-separation of all eight vitamin E isomers and Vitamin A,and is simple,sensitive,reliable and environmental protection.In order to avoid the time-consuming and complicated pre-treatment,accelerated solvent extraction(ASE)were applied to nut samples' pre-treatment.The extraction solvent can be used for inject just after constant volume,what makes the detection of Vitamin E isomers in nuts much easier,more accurate and sensitive.The above work then was used to investigate the occurrence and quantify the full isomeric composition of vitamin E(?-,?-,?-and ?-tocopherols and tocotrienols)in 3 kinds 210 samples by HPLC with fluorescence detection.The detection results were analyzed and concluded.A method to simultaneously determine Vitamin D2 and Vitamin D3 in milk powder,rice flour,mushroom powder,was also developed.The saponification solution was extracted with n-hexane and purified with SPE.The separation and detection of vitamin D forms was achieved by ultra performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).Isotopically internal standard was used to quantification.The method was verified sensitive,accurate and selective.
