Objective(1)Determine the extraction method of sapindus by orthogonal experimens,select the macroporous adsorption resin which can separate and purify the sapindus saponins;(2)Establish a HPLC method for simultaneous determination of sapindus saponins,and study on the HPLC fingerprint of sapindus and sapindus saponins;(3)Evaluate the surface activity and other properties of sapindus saponins.Method(1)To determine the optimal conditions,use orthogonal experimental,this paper discussed the influence of extraction time,solid-liquid ratio and ethanol concentration on extraction rate of sapindus saponins;With macroporous adsorption resin to Hederagenin retention as investigation index,choosed 9 types of macroporous adsorption resin for separation and purification.(2)Taking the Hederagenin as reference,the HPLC was applied to determine the content of Hederagenin.(3)Reference to the quality control of TCM injections in fingerprint technology,The similarity evaluation system for chromatographic fingerprint of TCM(2004 A edition)was used to evaluate the similarity of the 10 batches of sapindus and 10 batches of sapindus saponins for setting up sapindus and sapindus saponins of HPLC fingerprint.(4)determine the critical micelle concentration,surface tension and foaming properties of sapindus extract,and also study its influencing factors.Results(1)Through the orthogonal experiment,the optimal extraction process of sapindus saponins was: extraction solvent was 90% ethanol,solid-liquid ratio was 1:12,extracted 3 times,extracting time was 2.5 h,2.0 h,1.5h respectively,for this method,the dry extract yield was 4.17%and the content of Hederagenin was 2.315g;the optimal process of macroporous adsorption resin purification was: with DM130 macroporous adsorption resin,the saturated adsorption amount of sapindus saponins was 85.59(mg/g dry resin),elution rate was 93.1%;the concentration of sapindus extract was 0.5 g/m L(Hederagenin was 11.24 mg/m L),p H value was 5.3,the sample volume was 5.0 m L/g(adsorption liquid product: resin weight),the rate was 1 m L/min;the eluent was 95% ethanol and its p H value was 6.0,The elution rate was 1 m L/min.(2)Hederagenin in sapindus was linear in the ranged of 1.006ug-6.036?g,y = 495945x-14102,R2 = 0.9996,The average recoveries of Hederagenin were 99.94%,the methodological study showed that the method is simple,accurate and stable reliability.(3)The similarity evaluation system for chromatographic fingerprint of TCM(2004 A edition)was used to evaluate the similarity of the 10 batches of sapindus and 10 batches of sapindus saponins,the results showed that the sapindus and sapindus saponins fingerprints were feasilble,which could be used for index detection and quality control.(4)sapindus saponins had low critical micelle concentration and surface tension,and it had few effect by some factors(p H,temperature,salinity and water hardness)in a certain range;sapindus saponins had good foaming properties,good washing effect.Conclusion The extraction and purification technology of sapindus was feasible,and it can be used as sapindus' enlarge production process;the HPLC method was reliable for the determination of Hederagenin in sapindus;the fingerprint method was available,and can be used to evaluate the quality of the sapindus and sapindus saponins.sapindus saponins had strong surface activity,it can provide theory basis for the all-round development and research of sapindus source natural functional products.
