Chemiluminescence(CL)has become a more and more popular analytical technique owing to its high sensitivity,simple instrumentation,wide linear range,automatic,simplicity and rapidity.In this paper,a CL method is proposed for the determination of bovine serum albumin(BSA)and Hg2+ by using chemiluminescent probe.In detail,the thesis including such study as follows:1.A sensitive flow injection CL method is proposed for the determination of bovine serum albumin(BSA)using Copper(?)-Alizarin Red S(ARS)complex as an efficient chemiluminescent probe.The detection is based on the binding of the copper(?)-ARS complex to proteins and the catalytic activity of copper(?)-ARS in the Luminol-H2O2 CL system.Under the selected conditions,the CL intensity is linear with the concentration of BSA in the range of 5.0 x 10-11 to 1.0 x 10-9 mol·L-1.The detection limit was 2.0 x 10-11 mol·L-1.The method is successfully applied to the determination of protein in urine.2.A simple and sensitive aptamer-functioned AuNPs-based chemiluminescent probe for detecting mercury ion(Hg2+)in aqueous solution has been developed.The chemiluminescent probe relies upon the catalytic activity of unmodified AuNPs on the Luminol-H2O2 CL reaction,and the interaction of unmodified AuNPs with the aptamer.The unmodified AuNPs can effectively differentiate unstructured and folded aptamer.In the presence of Hgrig aptamer departs from the AuNPs as a result of the formation of G-quartet with Hg2+,leading to the increase of CL signal.By rationally varying the number of thymine(T)in single-strand oligonucleotides(ssDNA),the detection range could be tuned.Employing ssDNA with 14 thymine in the detecting system,a sensitive linear range for Hg2+ ions from 5.0 x 10-10 to 1.0 x 10-7 mol·L-1 and a detection limit of 2.1 x 10-10 mol·L-1 are obtained.Reducing the number of thymine to 10 and 6,the linear range is tuned.A low number of thymine makes the detection range narrow but the sensitivity high while a high number influences the detection limit but enlarges assay region.Besides,the probe also shows a good selectivity for Hg2+.3.We have developed a highly sensitive and selective chemiluminescence detection method for mercury ion by using a cysteine-AuNPs probe.This assay relies upon the thiols in the cysteine to form Au-S bonds with AuNPs,and the specific and strong interactions of thiols with mercury ion.In the presence of Hg2+,thiols tends to form Hgg+-S bonds,owing to their stronger affinity toward Hgg+ ions than that toward AuNPs,thereby resulting in the increase of CL signal.The concentration of Hg2+ can be determined by monitoring with the chemiluminescence detector.The limit of detection for Hg2+ is 8.5 x 10-11 mol·L-1(3a).This method exhibits excellent selectivity for Hg2+ over other metal ions.
