Recently,with the depletion of oil resources and increasing environmental pollution and the urgent need for sustainable development,rosin,as an environmentally friendly renewable resoure,has received more and more attention and been investigated extensively.In the present work,Rosin-based stationary phases of HPLC were prepared by using modified natural rosin as crosslinking agent.Then,they could isolate gastrodin from its derivatives.The structure and separation properties of rosin-based stationary phase were well investigated and the results are presented in this thesis.1.Ru131,Ru232 rosin-based microspheres and ester modified Ru131 microspheres were prepared,and characterized by using scattering particle size analysis,scanning electron microscope,specific surface area and pore structure analysis and thermogravimetric analysis.The results shows that the size of rosin-based polymer microspheres ranged from 3 to 8 ? m,which has abundant pore structure and good thermal stability at temperature below 300 ?.2.Ru131 were modified individually by methanol,ethylene glycol,Glycerol and glucose.And they were used as HPLC stationary phase to separate gastrodin and its derivatives,respectively.The influencing facts for separation of gastrodin and methylarbutin were carried out,including concentration of acetic acid,flow rate and column temperature.The results indicate that Ru131 was modified by methanol has the best resolution for gastrodin and its derivatives at 25? when methanol was used as moving phase and flow rate was 0.3 m L/min.And the best resolution reached 2.30.3.Ru232 was used as HPLC stationary phase to separate gastrodin and its derivatives,too.The influencing facts for separation of gastrodin and its derivatives were carried out,including the concentration of acetonitrile and acetic acid,flow rate and column temperature.The results show that gastrodin/phenolglucoside and gastrodin/methylarbutin mixtures could be effectively separated at 25 ? when methanol was used as moving phase,the concentration of acetic acid was 0.05% and flow rate was 0.3 m L/min.And the best resolution reached 1.54 and 2.73,respectively.Van't Hoff equation were used to calculate the thermodynamics parameters,including ?H,?S and ?G of gastrodin and its derivatives.4.Besides,Ru232 was used as HPLC stationary phase to isolated gastrodia from the aqueous extract of gastrodia elata.The results show the concentration of gastrodin increased from 39.28% to 82%.
