Zizyphus Jujuba cv.Jinsixiaozao has a long history of cultivation in China,it usually used as a diet to share.With rich nutrien,Zizyphus Jujuba cv.Jinsixiaozao,such as flavonoids,carbohydrates,cyclic adenosine monophosphate(cAMP)and other active substances.Zizyphus Jujuba cv.Jinsixiaozao has hypolipidemic,anxious,Anti-inflammatory,enhance immunity and other pharmacological effects,its development prospects are very broad.But the current processing of jujube are mainly dry products,the processing ways are very single,and the requirements of Zizyphus Jujuba cv.Jinsixiaozao are very high,some fruit with crack is often wasted.Due to environmental factors influence,jujube cultivation households tend to suffer huge economic losses.In recent years,many studies have shown that active substances in jujube can improve intestinal health(Liu Xiuling and Zhang Dai 2013;Liu Xinyu 2015),this study is particularly important for Zizyphus Jujuba cv.Jinsixiaozao of deep processing.In this paper,the fresh Zizyphus Jujuba cv.Jinsixiaozao of Laoling was used as the research object,low-molecule-weight sugars was extracted by the method of water extraction and alcohol precipitation.The fermentation low-molecule-weight sugars was obtained by fermentation.The effect of intestinal flora by low-molecule-weight sugars was tested by in vitro and in vivo experiments.Wish provide theoretical basis for Jujube health food development and application.The main contents of this thesis are as follows:1.Determine the best extraction of low-molecule-weight sugars water ratio of 1: 4,water extraction time of 3h and water extraction temperature of 80? by orthogonal experiment.Amount of optimum yeast was determined to be 1% by optimizing the fermentation conditions.Low-molecule-weight sugars(LMS)and fermented low-molecule-weight sugars(FLMS)from Zizyphus Jujuba cv.Jinsixiaozao were purified by membrane filtration and DEAE.The types and contents of low-molecule-weight sugars in jujube and the effects of fermentation on low-molecule-weight sugars content in jujube were determined by HPLC.Compared FLMS with LMS,the mixture of glucose and fructose was decreased by 2.98%,the content of sucrose decreased by 11.4% and the content of cane trisaccharide increased by24.6%.2.The effect of low-molecule-weight sugars from Zizyphus Jujuba cv.Jinsixiaozao on three kinds of bacteria growth in vitro was been studied.FLMS and LMS were added to the basal medium at different dose,OD value was measured at the best observation time.The results showed that a certain dose of FLMS and LMS can promote the growth of bifidobacterium(P<0.05),they inhibit the growth of E.Coli significant(P<0.05),and there areno significant influence on the growth of lactobacillus.3.The effects of low-molecule-weight sugars from Zizyphus Jujuba cv.Jinsixiaozao on the intestinal microflora of mice were studied.The mice in the experimental group were given different doses of low-molecule-weight sugars: unfermented low-molecule-weight sugars high dose group(WG)1.5g/(kg·d),unfermented low-molecule-weight sugars medium dose group(WZ)1.0g/(kg · d),unfermented low-molecule-weight sugars low dose group(WD)0.5g/(kg · d),fermented low-molecule-weight sugars high dose group(FG)1.5g/(kg · d),fermented low-molecule-weight sugars medium dose group(FZ)1.0g/(kg · d),fermented low-molecule-weight low dose group(FD)0.5g/(kg·d).The normal control group(CN)was fed with equal volume of distilled water for 14 days.After the last time,the feces of the mice were collected and stored in the refrigerator at-80?.The results show that the number of Ecol in the intestine of the mice was significantly lower than that of the normal control group(P<0.01).The effect of low-molecule-weight sugars group on the inhibition of Ecoli was lower than that of unfermented small molecule Obviously,FZ dose group had the best inhibitory effect.The low-molecule-weight sugars before and after fermentation could increase the number of bifidobacteria,and the effect of low dose group was the best.Jujube low-molecule-weight sugars can promote the growth of bifidobacteria in the intestinal tract of mice,they can inhibit the growth of Ecol and not achieve significant effect on the growth of Lactobacillus.4.Analysis of abundance and diversity of intestinal flora in mice,the change of weight in mice between 0 and 14 days was not obvious,indicating that the experimental group of oral dose had no effect on weight of mice.Compared with the CN group,WD,FG,FD group of mice after 14 days to fill the stomach,the size of the cecum became significantly larger,cecum total weight increased significantly(P<0.05),Wall of the cecum WG mice significantly higher than the CN group(P<0.05),compared with CN group,the other wall of the cecum experimental mice weight has no significant difference(P>0.05);Compared with the CN group,FG group the total weight of colon in mice increased significantly(P<0.05),FG and intestines of mice weight is significantly higher than the CN group(P<0.05).Each treatment group number of OTU was significantly higher than CN.FG,FZ,WG three groups was most than others.It is instruct that low-molecule-weight sugars can change the richness of gut microbes and is good for intestinal healthy.
