In the paper,we explore the extraction and purification process of anthocyanin from'Beilei' L.caerulea berries which are cultivated.Obtained anthocyanin extracts by optimal technological conditions.Extracts be used for the identification anthocyanins composition and antioxidant capacity are comparative analyzed.What'smore,Cy-3-glu were prepared,which is the most abundant in anthocyanin of 'Beilei' L.caerulea berries.We embedded Cy-3-glu into wall material aim to improve it's stability,and the stability of Cy-3-glu microcapsules were studied.Provide data support for 'Beilei' L.caerulea berries promotion and Cy-3-glu monomer application.The main results were as follows:(1)The effects of extraction technology including extracting agent concentration,so lvent type,extraction time,solid to liquid ratio on the content of Lonicera caerulea a nthocyanins were investigated using anthocyanins content as the evaluation index.The optimal technological conditions were determined via orthogonal experiment:Extraction solvent was 60%hydrochloric acid(0.1%)ethanol,solid-liquid ratio was 1:10 mg:mL,extraction time was 90 min.(2)Five macroporous resin(AB-8?X-5?D101?XAD7?NKA-9)was used to purify the obtained extracts,D 101 is the best one among those.The results showed that mass concentration is 1.0 mg/mL D101 macroporous resin of Lonicera edulis anthocyanin better purification effect,pH3.0,D101 macroporous resin adsorption capacity is relati-vely strong,the desorption timely using 60%ethanol solution.The total anthocyanin values is 353.35±0.79 mg/g,the purity increased from 4.76%to 35.43%.(3)HPLC-MS was used for the identification of anthocyanins composition,11 individual anthocyanins were found with cyanidin-3-glucoside(90.679%)as the major anthocyanin.Moreover,cyanidin3-(6'' acetylhexoside)(MS?491;MS2?287)was found for the first time in L.caerulea berry extracts.Additionally,antioxidant capacity and ABTS +·,DPPH radical scavenging activities of L.caerulea berry extracts,cyanidin-3-glucoside and VC were comparative analyzed,and the results were sorted as:cyanidin-3-glucoside>L.caerulea berry extract>VC.(4)Semi-preparative HPLC method was established for the preparation of Cy-3-glu with high purity from L.caerulea berry extracts.Cy-3-glu'purity reched 97.8%.The optimal technological conditions of preparation Cy-3-glu microcapsules were determined via orthogonal experiment:core materia:wall material=1:97,Soyaprotein:maltodextrin=2:8,Homogeneous time=8min.The microcapsule's target as follows:Embedding rate:92.17±1.87%,Microcapsule lation yield:78.42±1,23%,Solubility:76.21±2.14%,Moisture content:5.98±0.5%.(5)Study found that Cy-3-glu is very sensitive to sunshine and heat,but air's influence not soobvious.The results showed that the embedding treatment could significantly improve the sunshine,temperature,oxygen and digestive stability of Cy-3-glu.
