Lycium ruthenicum Murr.in Qing Hai province was researched in this paper.We mainly focused on ultrasonic extraction of polysaccharides from Lycium Ruthenicum Murr.,What's more,polysaccharides from Lycium Ruthenicum Murr.were purified?Then,we need to research the preliminary structure characterization and validation of antioxidant activity in vivo and in vitro experiments of purified polysaccharide from Lycium Ruthenicum Murr.1.The paper was mainly focused on ultrasonic extraction of polysaccharides from Lycium Ruthenicum Murr.,with the polysaccharide yield as the research object,single factor tests and orthogonal design method were applied to investigate the extraction conditions including material to liquid ratio,extraction time,extraction temperature and ultrasonic power.The results showed that the optimum extraction parameters were,extraction temperature 70 ?;extraction time 30 min;material to liquid ratio,1: 30(m/V,g:m L);ultrasonic power 200W;theyield of Lycium ruthenicum Murr.Polysaccharide yield could be 10.74 % under these condition.The rate is slightly higher than actual measured polysaccharide,believable results.2.The purification polysaccharides from Lycium Ruthenicum Murr..It deproteinized by Three chloroacetic acid,then,the curde polysaccharides was purified by ultrafiltration,macroporous adsorption resin,polyamide chromatography elution,fractional precipitation,DEAE-52 cellulose and dialysis water purification,and the high performance liquid chromatography(HPLC),On the elution with DEAE-52 cellulose column high performance liquid as a result,there is almost no difference.Results show that after fractional precipitation,the polysaccharides are by the LPR-60,LPR-70,LPR-80,LPR-90 four polysaccharide composition,content of 69.27%,76.07%,78.11% and 74.43% respectively.After DEAE-52 cellulose column elution,and the preparation of high performance liquid results almost no discrepancy,the experimental results concluded that the polysaccharides by the LPR-80-1,LPR-80-2,LPR-80-3 and LPR-80-4 four polysaccharide composition,content of 76.68%,78.59%,78.54% and 78.59% respectively.Select polysaccharide components LPR-80-2 for subsequent trials.3.This paper makes a preliminary analysis about the structure of polysaccharide of LPR-80-2 by ultraviolet spectrum,infrared spectrum,capillary electrophoresis,scanning electron microscopy(SEM),atomic force microscopy,mass spectrometry.Results of UV test show that the polysaccharide component is not obvious absorption peak at 260-280 nm,it shows that the polysaccharide samples do not contain proteins and nucleic acids and other biological macromolecules;Infrared spectrum analysis showed that polysaccharides from Lycium Ruthenicum Murr.has the characteristic absorption peaks of the polysaccharide,and contain pyran ring structure;And capillary electrophoresis results found that the polysaccharide components composed of rhamnose,arabinose,glucose and galactose sugar four kinds of monosaccharide composition,and its composition mole ratio is: Rha: Ara: Glc: Gal = 0.3:2.2:1.0:1.5;Atomic force microscope scanning showed that the polysaccharide molecules between molecules are close,circle shaped stacking structure;Scanning electron microscopy(SEM)show that the polysaccharide surface present a rough laminar structure.4.To study the antioxidant activity of crude polysaccharides from Lycium Ruthenicum Murr.And the purified polysaccharide scavenging DPPH free radicals,superoxide anion radicals and hydroxyl radicals in vitro.The result showed that crude polysaccharides and the purified polysaccharide from Lycium Ruthenicum Murr.have the ability of scavenging free radicals,they all show certain antioxidant activity,and the ability of the purified polysaccharide slightly higher than the crude polysaccharide of scavenging free radicals,and the effects of free radicals in the certain concentration range polysaccharide solution has certain concentration-response relationship,with the increase of concentration,the ability of scavenging free radicals also increases,the study also found that the polysaccharide samples hydroxyl free radical scavenging ability is significant,with the increase of concentration,more and more close to the Vc's ability to remove active oxygen free radicals.The study of polysaccharides from Lycium Ruthenicum Murr.is about liver protective effect of cadmium poisoning mice,the liver MDA content and antioxidant enzymes in the CAT and the content of NO and SOD content as an index,the results showed that the black fruit of Chinese polysaccharides could reduce MDA content,SOD and CAT activity increased,the content of NO,confirmed the black fruit of Chinese wolfberry has certain antioxidant effect in vivo,to cadmium poisoning in mice liver has a protective effect.
