Ganoderma lingzhi is a kind of precious medicinal fungus. As a new medicine Ganoderma lingzhi spore powder is widely used in health product. The strains,cultivation methods,management model, and other factors will affect the quality of Ganoderma lingzhi spore powder during the cultivation process. In addition, the broken process and extraction technology will bring about changes to the quality of Ganoderma lingzhi spore powder. However, there are no systematic studies on the differences of the active components in Ganoderma lingzhi spore powder products from different sources. Therefore, the analysis method of polysaccharides and triterpenes in Ganoderma lingzhi spore powder was further optimized in this paper based on the previous work of the laboratory. The components including polysaccharides, triterpenoid,nucleoside, sterol and other active ingredients in Ganoderma lingzhi spore powder products from different sources were analyzed and compared to explore their quality differences.Optimization the determination method of polysaccharide content in Ganoderma lingzhi spore, the optimal conditions were to be material to liquid ratio 1:50, solid to liquid ratio 1:10,precipitation time 9 hours, centrifugation 10000 r / min. Using the new method, it was found that there were significant differences in the contents of polysaccharides in Ganoderma lingzhi spore powder. According to HPLC fingerprint, there is a obvious difference among peak number and peak area, using the ion chromatography-integrated to determinate the monosaccharide composition in polysaccharide, The proportion of glucose in monosaccharide composition is obviously different. Therefore, from the perspective of the active ingredient polysaccharide,different sources of Ganoderma lingzhi spore powder products are significantly different.The quantitative analysis multi-components by single mark (QAMS) method for the determination of triterpenes in Ganoderma lingzhi spore powder was established. The method can be used to determine the content of 12 Ganoerma lingzhi triterpenes in Ganoderma lingzhi spore powder at the same time. The RSD of precision, stability, reproducibility and recovery rate were all less than 5%, which indicated that the method was reliable, provided a new idea for the determination of triterpenes in Ganoderma lingzhi spore powder. The results of this method showed that there were also significant differences in triterpenes species and content between different sources of Ganoderma lingzhi spore powder products.The results showed that there were significant differences in the content of nucleosides,ergosterol, erythrothioneine, choline in different sources of Ganoderma lingzhi spore powder products through the methods established before. At the same time, the comparison of microscopic identification, broken wall rate, water content and oil content were also carried out.The results show that the breaking ratio is higher than 90%, the oil content is higher than 20%,and the water content is less than 7%.Through the analysis of Ganoderma lingzhi spore powder on the market, this paper laid the foundation for the establishment of quality control standard of Ganoderma lingzhi spore powder.
