Preparation Of Poly (GMA-co-EDMA) Monolithic Column And Expand Its Application

The monolithic column has become a rapidly burgeoning field in recent years.Compared with the traditional packed column,the advantages of easy preparation,low cost,high-resolution,good stability as well as good hydrodynamic characteristic make monolithic material an ideal support for the application field of chromatography.In spite of this,a riper,perfect preparation technology to the monolithic column has not formed,which need be studied deeply and deflected step by step.In this paper,in situ polymerization method was employed to prepare poly(GMA-co-EDMA).The glycidyl methacrylate(GMA)be chosen as functional monomer,,and ethyleneglycol dimethacrylate(EDMA)be chosen as cross-linking agent.The effects of key factors such as porogen on porous structure of poly(GMA-co-EDMA)was investigated by single factor method.Then,the organic polymer monolithic column with good shape,high efficiency and good permeability was obtained.Furthermore,six groups of different compounds were separated successfully on the monolithic column and the resolution was larger than 1.5,which was indicating that the poly(GMA-co-EDMA)was suitable for separation and analysis of small molecule mixtures.In addition,poly(GMA-EDMA-DEAE)weak anion-exchange monolithic column was achieved by modification of epoxide groups of the poly(GMA-co-EDMA),which reacted with diethylamine(DEAE).The influences of some variables such as pH,temperature,buffer concentration and BSA concentration on the binding capacity of BSA and breakthrough curves of the poly(GMA-EDMA-DEAE)were determined.After that,the D,L-tryptophan was separated successfully and the resolution was 1.59 on perpared BSA affinity monolithic column..The results confirmed the excellent separation efficiency of the prepared column.In this paper,the separation of small molecules and chiral drugs were achieved by prepared monolithic columns,which provided a new idea and new method for the application of organic polymer monolithic column.Therefore,the experimental research was significant and valuable.
