Epimedium is a Chinese traditional medicine commonly used in clinical Chinese medicine,which was highly favored by ancient physicians.Modern pharmacological studies had made a reasonable explanation for its many of the traditional efficacy.Its chemical composition had been more comprehensive in-depth study.The chemical components of Epimedium are inclouding flavonoids,alkaloids,and polysaccharides.Currently,Preparation process of Epimedium's active ingredient had two pionts to attention worthly.First,Epimedium flavonoids or polysaccharides,such as a single active ingredient preparation process is more than comprehensive utilization of a variety of active ingredients;second,a byproduct of the preparation of epimedium polysaccharide contains a lot of epimedium flavonoids and alkaloids,and now basically as waste was discharged,not only wasteful,but also increase the discharge of pollutants,and the recycling processes had not been reported.To solve the these issues,The focus on effective methods of recycling Epimedium flavonoids,alkaloids,from preparation of epimedium polysaccharide waste.The establishment of comprehensive utilization of the preparation process of Epimedium variety of ingredients.In this study,The ultrafiltration exudate of Epimedium extract flowed through the cation column firstly,then the Effluent flowed through the anion column.The result shows that this method can recycle two kinds of effective component which were inclouding Epimedium flavonoids and Epimedium alkaloids.Yet its selective separation effect was still not ideal,there was a part of the mixture.of the two kinds.Second,The ultrafiltration exudate of Epimedium extract flowed through the anion column firstly,then the Effluent flowed through the cation column.The results reveals Epimedium flavonoids can be effective adsorption by anionic resin,alkaloid components outflow,Alkaloids of the residual in the anion column can be eluted then effluent and alkaloids eluent flowed through the anion column Alkaloids can also be effective adsorption and elution.The Separation method Can achieve the purpose of selective separationOn the basis of the above experiments,select the optimum purifying process of Epimedium flavonoids in Ultrafiltration exudative fluid by different eluents on the anion exchang resin of strong base.Different eluents such as ethanol and hydrochloric acid were researched,with elution rate of Icariin as the indexes and determined byHPLC.The results interperts 70%ethanol and 1%hydrochloric acid elution better:The former the icariin peak area of 4.2 mAU*min,the latter was 1.5 mAU*min,Through Ethanoland hydrochloric acid are used in conjunction and orthogonal experiments,the optimal condition was that the aleoholeon centration was 70%,hydrochloric acid content of 0.5%the peak area of icariin was 6.6 mAU*min,In addition,with the above process research,Epimedium alkaloids and flavonoids detection analysis methods also were did a series of studies.The analytical methods which are including high performance liquid chromatography and thin-layer chromatography were established to detect flavonoids and alkaloids in Epimedium.The acid-base titration was also adopted to detect total alkaloids content.The Epimedium flavonoids detection analysis methods use HPLC gradient elution,the map which was experimentally determined gradient elution conditions shows the icariin peak shape and resolution are relatively satisfactory.The method can be used in conjunction with the above process.
