5a-reductase inhibitors can retard the biosynthesis of dihydrotestosterone to improve or alleviate benign prostatic hyperplasia.They are applied for the treatment of benign prostatic hyperplasia extensively.Bee Pollen has a variety of nutrients and physical activity,so it has a unique effect in the treatment of prostate disease.Firstly,spectrophotometric method were used to establish 5a-reductase screening model in vitro:testosterone 75?mol/L,NADPH 60?mol/L,enzyme extract 66 pg,DTT 1 mmol/L,pH6.5 sodium phosphate buffer 20 mmol/L,37 ? for 4 min.Under this condition,five determination of the reaction tube and a blank tube reaction rate:5.35±0.11 ?M/min,RSD2.06%and 2.85±0.15 ?M/min,RSD5.26%.The inhibiton of 12 kinds of bee pollen different extracts on 5 a-reductase are tested and their efficacy order is supercritical fluid extract from bee pollen>the alcohol extract from bee pollen>the water extract from bee pollen;then the inhibiton of 12 kinds of different concentrations of the supercritical extract of bee pollen on 5 a-reductase are tested,It shows that the inhibition of the coquelicot bee pollen,schisandra bee pollen,corn bee pollen,sun:flower bee pollen on the activity of 5a-reductase is stronger than the other bee pollen.The IC50 value is about:the coquelicot bee pollen,145 ?g/mL;schisandra bee pollen,105 ?g/mL;corn bee pollen,145 p.g/mL;sunflower bee pollen,135 ?g/mL and close to the IC50 value of the positive control finasteride.Supercritical fluid extraction of plant sterols in four bee pollen were analyzed by GC-MS and found that schisandra bee pollen has the most types of plant sterols,coquelicot bee pollen and corn bee pollen have more types of plant sterols,and sunflower bee pollen at least.Campesterol,stigmasterol,(3-sitosterol and ?7-avenasterol were present in bee pollen.Supercritical fluid extraction of plant sterols in schisandra bee pollen was evaluated in this study,the effects of main operating parameters including extraction pressure,temperature and time on the extraction yield of plant sterols were studied.The maximum extraction yield of 8.69 mg/100g was achieved at a temperature of 53 ?,a pressure of 38Mpa,a time of 107 min.
