The phenomenon of radial heterogeneity in the liquid chromatography column is ubiquitous,which causes the sample band broadening during the process of solute migration,leading to the widening of the spectrum and the poor peak symmetry(the front of the peak or the tail),and as a result affects the separation efficiency of the column.In particular,for the preparative chromatography,the larger the radial dimension of the column,the greater the negative effect of the mobile phase radial heterogeneity.Therefore,in this study,from the internal structure of the column,we changed the migration path of the mobile phase in the column,weakened the influence of radial heterogeneity and improved the separation efficiency through a series of embedded structure liquid chromatography column design.First of all,FLUENT hydrodynamics software was used to simulate the migration trajectories of the mobile phase in the ring-embedded chromatographic column.The mechanism of the effect of the nested structure column on the chromatographic process was expounded from the theoretical point of view.And then used the AE(adobe after effect)software to completely demonstrate and intuitively show the fluid flow process in the ideal structure.Then,a series of embedded structure columns were designed,and the influence of the embedded structure on the chromatographic behavior was revealed by experiments.The optimal structure design parameters were obtained.Certain research results have been obtained in this study.Firstly,the influence of the embedded structure column on the chromatographic process was explored from the theoretical point of view by using the FLUENT software to simulate and calculation.The simulation results showed that the ring flow embedded column can effectively change the migration trajectory of the mobile phase in the column,optimize the distribution of the mobile phase and improve the chromatographic behavior.Under the condition of simulated experiment,the ring embedded chromatographic column whose effective radial distance of the column was 80%of the column diameter showed relatively good result.Under different column length conditions,the number of theoretical plates increased compared with the traditional columns,and the tailing factors were reduced,the symmetry of the peaks was better,and the column separation efficiency was higher.Secondly,the actual chromatographic experiments showed that different segmentation ratios had a great effect on the chromatographic behavior,and there was a better segmentation ratio to improve the column efficiency.The segmentation ratio affected the flow state of the mobile phase inside the column and the peak shape.The best proportion of a obtained in this study was 1:2:2:1.Lastly,the simulation experiment has certain guidance on the chromatographic experiment.The chromatographic experiment also verifies the speculation of the simulation experiment.The research of the embedded structure has laid a theoretical foundation for the future design of the new column structure,and provides a new development direction.
