Akebia Trifoliata is the woody vines and half long green twinning plant.Akebia Trifoliata mainly contains flavonoids,polyphenols,oleanolic acid,linoleic acid,micrielement,and so on.Akebia Trifoliata and its extractives are applied in pharmaceuticals,food,and cosmetics.In this paper,Akebia Trifoliata from western Hunan region having been regarded as the raw materials,determination of total triterpenes in fruit of Akebia Trifoliate peel by ultraviolet and the process conditions of preparation total triterpenes by Akebia Trifoliata peel hydrolysis were investigated.Adsorption kinetics and thermodynamics analysis of macroporous resin for total triterpenes and the separation technology of response surface optimization of total triterpenes by macroporous resin were studied.In order to establish a method of determining the content of total triterpenes in the fruit of Akebia Trifoliate peel,the optimization of chromogenic conditions and investigation of methodology were carried out with Vanillin-perchloric acid as color development reagent and 545 nm as the maximum absorption wavelength.UV spectrophotometry method measuring content of total triterpenes is compared with the HPLC.Optimal reactive conditions for 0.20 mL sample solution are obtained as follows: color development reagents 0.30 mL 5% vanillin and 0.70 mL perchloric acid,temperature 70 ?,reaction time 15 min.The average recovery of total triterpenes was 101.1%,and the RSD value is 1.2%.The UV spectrometric method is rapid,accurate,stable and reproducible and can be used for determining content of total triterpenes in the fruit of Akebia Trifoliate Peel.In addition,measuring results in this method and HPLC is consistent.Total triterpenes in Akebia Trifoliata peel was extracted by ultrasonic.The effects of pH,extraction time and extraction temperature on the extraction rate of total triterpenes were investigated through single factor experiment.The results showed that the optimized extraction parameters: pH = 5.6,extraction temperature of 71 ?,extraction time for 103 min,optimum conditions,the extraction of total triterpenes rate for 97.2%.The separation of Akebia Trifoliata peel total triterpenes with seven commercial macroreticular resins were studied.The results showed that XDA-1 is optimal resin for the separation of Akebia Trifoliata peel total triterpenes.The research aimed to study the kinetics and the thermodynamics of adsorpting total triterpenes by XDA-1 macroporous resin.Some adsorption characteristics was investigated based on the pseudo-first-order,pseudo-second-order model,and thermodynamic parameters for the adsorption of total triterpenes were determined using the Van't Hoff equation.The results of kinetics study showed that adsorption of total triterpenes by XDA-1 macroporous resin conformed to the pseudo-second-order kinetics equation.The results of thermodynamics study demonstrated that adsorption of total triterpenes by XDA-1 macroporous resin conformed to Freundlich adsorption isotherm equation.The enthalpy changes ?H<0 indicated that the absorption was an exotherm process,and |?H|<43 kJ/mol demonstrated that the absorption process was physical adsorption.Meanwhile,the free energy changes ?G<0 indicated that the absorption was a spontaneous process.The entropy changes ?S>0 demonstrated that the absorption process was the process of entropy increase.On the basis of single factor experiments,influence of solid-liquid ratio,adsorption temperature and adsorption time on the total triterpenes extraction yield were investigated by response surface optimization.The experimental results showed that extraction yield of total triterpenes is 63.53% in solid-liquid ratio for 1:8 g/mL,40 ?temperature adsorption 315 min.Predictive value of model on total triterpenes extraction yield was 58.17%,it was anastomotic compare with result of combination experiment.
