Studies On Extraction,Separation,Purification And Biological Activity Of Chenopodium Quinoa Saponins

Chenopodiumquinoa saponins is considered one of the important effective components in chenopodium quinoa willd.But the structure-function relationship of chenopodium quinoa saponins has not been clear.In this study,chenopodium quinoa seed were used as raw material to extract and purify chenopodium quinoa saponins,and its antioxidant activity and antibacterial activity were researched.The specific research contents and results are as follows:(1)The parameters of reflux extraction and ultrasonic assisted extraction are selected and optimized to extract chenopodium quinoa saponins.The extraction ratio 1.763g/100g of reflux extraction,and the optimal technological condition are as following:feed liquid ratio 1:32,ethanol concentration 89%,extracting 94 min at 83 ?.The extraction ratio 1.892g/100g of ultrasonic assisted extraction,and the optimal technological condition are as following:feed liquid ratio 1:31,ethanol concentration 94%,extracting 95min at 51?.Results indicate that the effect of ultrasonic assisted extraction was more obvious.(2)The optimum parameters,concetration 4.0mg/mL,pH value 5,eluent velocity l.OmL/min,eluent concentration 90%by ethanol,were determined by the static/dynamic adsorption and desorption experiments of D101 macroporous adsorption resin.The purity of chenopodium quinoa saponins is 36.2%.(3)The purifying of chenopodium quinoa saponins A and B was respectively obtained by silica gel column chromatography and preparative high performance liquid chromatography,the purity was 86.2%and 89.5%.(4)Study on antioxidant activity of chenopodium quinoa saponins in vitro.The results show that chenopodium quinoa saponins can scavenge DPPH·,OH·,O2-· at a certain concentration,and the chenopodium quinoa saponins has strong ability to restore Fe3+.It indicating that the antioxidant activity of chenopodium quinoa saponins is strong.(5)The antimicrobial activity of chenopodium quinoa saponins was studied.The results showed that chenopodium quinoa saponins is a broad spectrum bacteriostatic activity materia,which has certain inhibitory effect on some gram-positive bacteria,gram-negative bacteria,cocci,bacillus and fungi.At the same time,the test results show that chenopodium quinoa saponins is sensitive to staphylococcus aureus,the minimum inhibitory concentration is 0.625 mg/ml.0.625 mg/ml.
