Asparagus old stems are a byproduct of asparagus processing,containing polysaccharides,saponins,polyphenols and other active substances.The development of it is of great significance.Early work shows that asparagus powder can improve sleep,with the potential to regulate brain function.Therefore,the aim of this study was to investigate the effect of asparagus juice(AEAS)on memory impairment in mice,to determine whether it had the potential to interfere with mild cognitive impairment(MIC),and to investigate the role of asparagus juice in regulating intestinal flora.In this work,the content of nutrient and active ingredients in asparagus juice(AEAS)was determined by colorimetry and liquid chromatography.The results showed that AEAS with the solid content 12.5% contains polysaccharides,total saponins,polyphenols and amino acids and other active substances.Polysaccharide content of up to 50 mg/m L,followed by total saponins and low level of polyphenols.Polysaccharides consist mainly of arabinose,galactose,galactose,glucose,xylose and mannose at a ratio of 0.036 : 0.14 : 0.39 : 1 : 0.038 : 0.21.Amino acids contain large amounts of gamma-aminobutyric acid,more than 1 mg/mL.Then 60 healthy ICR mice(4 weeks old)were randomly divided into six groups(the sample recomputed with solid matter): control group(CT),model group(SC),positive control group(DON),low dose of AEAS group(LS,0.2 g /kg weight),middle dose of AEAS Group(MS,1 g /kg weight)and high dose of AEAS group(HS,2 g /kg weight).The respiration rate,calorie and activity of mice were measured by animal monitoring system.The effect of asparagus juice on memory impairment was investigated by mouse behavioral experiment.The results showed that LS group and MS group could significantly increase the activity of mice at night,which were increased by 90.9% and 112.8%,respectively.In the behavioral experiment,the open field experiment showed that AEAS did not affect the mouse independent activity and anxiety caused by mice and other negative effects.The new object recognition experiment showed that MS group could significantly improve the discrimination index of memory impairment mice(P<0.05).In the Y-maze,the spontaneous alternation of the MS group was significantly different from the model group(P<0.05).The MS group could significantly improve the latency of the mice(P<0.05)and the MS group had a significant improvement in the target quadrant duration(P<0.05)during the test period.The contents of acetylcholine(ACh)and acetylcholinesterase(AChE)in the hippocampus were measured.The levels of malondialdehyde(MDA),superoxide(SOD),catalase(CAT)and total antioxidant capacity(T-AOC)were measured.Expression of memory-related genes in hippocampus was determined.The results showed that ACh in MS group was significantly increased(P<0.05)and AEAS could significantly inhibit AChE activity(P <0.05).The levels of brain-derived neurotrophic factor(BDNF),cyclic adenosine monophosphate effector binding protein(CREB)and nerve growth factor(NGF)in the MS group were significantly higher than those in the SC group(P<0.05).AEAS can improve CAT and T-AOC activity in brain and serum(P<0.05).Finally,we explored the effect of asparagus juice on intestinal flora in mice.The results show: AEAS significantly decreased colonicacetic acid and increased lactic acid and butyric acid(P<0.05).AEAS significantly increased fecal Akkermansia,Allobaculum,Lachnoclostridium,Marvinbryantia,Turicibacter and Parabaceteroides,and decreased Staphylococcus,Lachnospiraceae and Ruminococcaceae.Compared with CT group and SC group,LS group could increase the expression of IL-10 and IL-22 genes in the infarcted group(P<0.05).The inflammation-related genes in the jejunum were not significantly changed.The correlation analysis shows that butyric acid was positively correlated with Akkermansia and Marvinbryantia,but was positively correlated with ACh(P = 0.008,R2 = 0.6).Shannon index was significantly negatively correlated with AChE(P = 0.001,R2 = 0.728).Staphylococcus had a significant positive correlation with AChE activity(P = 0.014,R2 = 0.567).Marvinbryantia had a significant negative correlation with AChE activity(P = 0.025,R2 = 0.641),while Parabacteroides and Akkermansia were positively correlated with the time of the target quadrant(water maze test)(P = 0.016,R2 = 0.497;P = 0.05,R2 = 0.409).The results of the final study showed that AEAS can improve the memory impairment induced by scopolamine in mice and change the community structure of intestinal flora.Therefore,AEAS has the ability to interfere with mild cognitive impairment.
