Peony seed oil is rich in unsaturated fatty acids which are mainly ?-linolenic acid,linoleic acid and oleic acid and plays an important role in human health.Lead is a common toxic heavy metal in the environment,which can not be degraded.It accumulates in the human body through atmosphere,water and food,impairs human memory and cognitive ability,thus influencing children's intellectual development,and making human health damaged.In this study,the repairing effect of peony seed oil on lead-induced learning and memory impairment in rats was investigated for developing an effective method of food intervention to prevent and repair the lead poison.On this basis,the peony seed oil buccal tablet was prepared with peony seed oil,which has the advantages of more stable chemical quality,convenient storage and carrying,and accurate measurement.Method:1.Animal grouping and administration:32 healthy weaning SD rats were selected,each weighing about 200 g.They were randomly divided into normal group,peony seed oil group,lead exposed group and lead exposed group + peony seed oil group which individual group contain 8 rats.The animals in the normal group were fed with deionized water and standard feed(AIN93G)while the other treatment groups drink up 250 mg/L lead acetate solution(Pb AC)and take the food with peony seed oil in the ratio of 9 g/100g?2.The repairing effect of peony seed oil on lead induced learning and memory impairment was studied by Y maze food reward experiment and Morris circular water maze experiment with the key parameters recorded by software,such as the path of the four groups of rats in Y maze arm,the number of times of entering and leaving maze arm and the movement track of rats in Morris water maze experiment.3.Peony seed oil buccal tablets were prepared with peony seed oil and ?-cyclodextrin.The chemical conditions was optimized by controlling variable method with the reaction time,reaction temperature and alcohol oil mixture ratio which influenced the embedding rate of peony seed oil buccal tablets.Result:1.In the process of behavior experiment,compared with the lead exposed group,the result show that the number of times that the former went in and out of each maze arm in Y maze experiment remained unchanged for lead exposed group + peony seed oil group.In the navigation experiment,compared with the lead exposed rats,the movement track of the rats who often took peony seed oil was more convergent,and they could find the platform faster.In the experiment of space exploration,in 120 s,the frequency of SD rats in the lead exposed group + peony seed oil group swam through the original platform was 2-3 times than that in the lead exposed group.The former stayed in quadrant I(platform location)longer,and had a deeper memory of the original platform location.2.In the experiment of embedding rate of peony seed oil,by optimizing the factors such as reaction temperature,reaction time and the ratio of alcohol and oil,the results show that the embedding rate of peony seed oil buccal tablet can reach about 36% under the conditions of the ratio of alcohol to oil 15:1,reaction temperature 40 ?,reaction time 60 min.Conclusion:Based on the behavioral experiment,it was found that the rats in the lead exposed group performed the worst in the behavioral experiment,compared with the rats in the lead exposed group fed with peony seed oil,indicating a better intervening effect of peony seed oil on the memory damage caused by lead.These results may provide a new sight for the intervention of memory damage caused by lead.In the preparation of peony seed oil buccal tablets,there are many factors that affect the embedding rate of buccal tablets.In this paper,the main influencing factors such as reaction time,reaction temperature and alcohol oil mixture ratio on the embedding rate of buccal tablets are carried out to confirm the best reaction conditions.
