In this experiment, obese rat model was established by high fat diet, two kinds of intervention programs of natural bran and GABA-enrich bran were applied for study the effect of high fat diet on rats weight, blood glucose, blood lipid, oxidative stress, liver function and using the real-time fluorescence quantitative PCR study the intervention methods on the rat fat metabolism, glucose metabolism, oxidative stress and liver function related gene expression level parameters.Firstly, a new kind of increase content of GABA enrichment in wheat bran by adding glutamate, by HPLC on enrichment results were determined, the results show that the content of GABA increased from 38.24 mg/100 g to 248.76 mg/100 g, increased by 6 times or more. The antioxidant activity analysis showed that GABA-enrich bran antioxidant capacity (DPPH. removal rate and ABTS. removal rate) was significantly higher than that of natural wheat bran,which may be in the process of the GABA rich bran culture promoted the transformation and synthesis induced by polyphenols.Secondly, obese rat model was established by high fat diet, the effect of the intervention on rats blood glucose, fat metabolism, glucose metabolism, oxidative stress and liver function level of the structure under high fat diet, and observe the morphological changes of liver tissue at the same time under different feeding conditions. The results showed that: compared with the normal control group and high fat group,natural bran and GABA-enrich bran has rich natural slimming effect, can reduce weight, reduce the serum insulin level, total cholesterol and triglyceride content, increase the total antioxidant capacity and the removal of free radicals, inhibition of liver tissue lesions.Finally, the gene expressions responding for live lipids metabolism, oxidative stress,live function were examined for revealing the mechanism.,in order to reveal the mechanism of natural bran and GABA-enrich bran. The expression of the metabolism and glucose metabolism in the liver of fat related genes such as ACC, FAS, Acoxl, SREBP, Acly, Fadsl,Gpam, Dgatl, GYG1, G6PC1, GS2, PEPCK, Insigl, Insig2, etc., can effectively inhibit the synthesis of the influence of body fatty acid, glycerin three greases and cholesterol. The results of the study also found that: GABA-enrich bran intervention by enhancing Acoxl expression of lipid metabolism related genes, can significantly improve the ability of fat?-oxidation, so as to further improve the blood lipid in obese rats, promote anal glucose in liver tissue transferred into glycogen, blood glucose level in serum decreased, it also preliminarily reveals the internal mechanism GABA rich bran regulate lipid metabolism and glucose metabolism disorders. At the same time, the changes of oxidative stress in the liver transcription level was also studied through the determination of related genes VEGF,HMOX1, IGBFP1, PM1, GAPD on oxidative stress, and liver function related genes Gsta2,Gsta3, Gstp1, Gstm2, Nqol, Gclc, Gclm expression levels, found that the intervention can effectively improve the level of oxidative stress obese rats, remove excess free radicals,improve the total antioxidant capacity, improving liver metabolism function of the organism, which provides a scientific basis for improving liver metabolism of wheat bran is rich in GABA, provides some theoretical guidance for the development and function of the effective utilization of wheat bran.
