Topic Basis: Shanxi aged vinegar(SAV)has a variety of medical functions such as resisting oxidation,decreasing blood sugar and blood lipids,etc.,with special taste and abundant nutritional value.In recent years,lots of work had been done,the nutritive compositions(amino acids,organic acids,etc.)in SAV were clarified basically.However,the types and structures that have certain features to use remain largely unknown.During the aging,besides water volatile and increase of acidity,are there any other chemistry transformation to reflect their quality Compared with other types of vinegar that were brewed by cereal,are there any other compositions with specific benefits? Thus,based on the recent research to clarify the characteristics of the SAV functional componentby1 H NMRand HPLC finger-printmethod and to improve its quality control methodIn view of the functional component were worth further research.Purpose: SAV fingerprinting was built up to evaluate the different kinds of vinegar and Shanxi vinegar with different aging time,and illustrated the characteristic of functional components of SAV;The extracts of ethyl acetate was evaluated in the bioactivity;The chemical difference of SAV with different aging time was evaluated,which will make a differences.Methods: The ~1H NMRfingerprinting of SVA was optimized and applied to compare the SVA with different aging time.HPLCfingerprinting was built and analyzed the functional components of SAV.Reference substance was separated and applied to distinguish chromatographic peak of fingerprinting that was used to analyze the saled SAV and the other types of vinegar,and then confirmed the characteristic of functional components of SAV and explored the bioactivity.Based on the chemical difference between SAV and SMV,~1H NMR metabonomics technology was used to evaluate the difference body metabolism of SD rats.Results:(1)Through investigating the test methods of SAV ~1H NMR,~1H NMRfingerprinting was optimized and built,and applied to the compared results of SAV with different aging time.In the SAV,the content of most of compounds is higher than SMV,but SAV/SMV of difference compounds is different.The various compounds presented the dynamics changing process in the process of aging.(2)According to the functional compositions in the SAV,the HPLC fingerprinting was built and identified 13 chromatographic peaks,concluding p-hydroxybenzoic acid,DHCA,dihydrocaffeic acid,vanillic acid,protocatechuate,ferulic acid,dihydroferulic acid,5-hydroxymethylfurfural,trans-p-hydroxycinnamic acid,ligustrazine,syringic acid,caffeic acid,catechin,L-epicatechin.Fingerprinting can be used to the homogeneity evaluation of saled SAV and distinguish of other vinegar(rice vinegar,balsamic vinegar,series of vinegar,etc.).Preliminary activity experiment results show that 0.4 mg/mL the extracts of SVA acetic ether can prolong the life of nematodes.200 ?g·L-1the extracts have no nerve protection and neurotoxicity to PC12 cell.(3)On the basis of chemical difference between SAV ahd SMV,a NMR metabonomicstechnologycombined with multivariate statistical analysis was conducted to compare the differenceof the two types of vinegar in vivo.The key metabolites in the serum that contributed to the separation of the vinegar-given rats from the control rats were decline of 3-HB,methionine,glutamate,glutamine and acetone,as well as elevation of lactate,acetate,alanine,threonine,?-glucose and ?-glucose.The rats in SAV group contained more acetate and lactate,as well as less citrate,histidine,serine,threonine and ?-glucose compared with the SMV group.Conclusion: Building up the SAV ~1H NMRfingerprinting can be used to evaluate the Shanxi vinegar with difference aging time.The HPLC fingerprinting has excellent stability,high precision,good repeatability,which can be used to the homogeneity evaluation of saled SAV and distinguish of other vinegar.The functional components of SAV werevanillic acid,protocatechuate,dihydroferulic acid,ligustrazine,syringic acid,caffeic acid.After giving the vinegar,the metabolic pathways of the body metabolism of SD rats are amino acid metabolism,energy metabolism and intestinal flora metabolism.Compared with the chemical differences,SAV with difference aging time has little influence to the body metabolism of SD rats.
