Chitosan,the only basic polysaccharide in natural polysaccharide,is an natural polymer compound less than cellulose.Due to its good ability of weight reducing,it is widely applied to functional food.Chitosan as an pharmaceutical excipients have been added in the Chinese Pharmacopoeia,but there still have not standard method for its quantitation.Thus,to etablish chitosan quantitative method is helpful to etablish its quality standard and to guarantee the related product stable.Anionic surfactant,with good emulsification,foaming,dispersion,decontamination,and so on,have been widely used in daily life,textile,medicine,mining and construction industries.However,a large number of industrial wastewater and domestic wastewater which contain anionic surfactant,is discharged into aquatic environment without any treatment.It will kill aquatic organisms,cause massive blooms and red tide phenomenon.Therefore,the detection of the content of anionic surfactants in water is of great significance to the protection of water environment.Since 1990,resonance Rayleigh scattering(RRS)technique,as a new analysis method,has been successfully applied to analyze the biomacromolecules(nucleic acids and proteins),anionic surfactant,dye,drug,etc.Scholars have been found that ions of opposite charge would form binary or multiple ion association by electrostatic force,hydrophobic function and charge transfer effect.Resonance Rayleigh scattering intensities are greatly enhanced and the increased intensities are proportional to the concentration of material,which realize the aim of quantitative analysis on various materials.With its advantages of easy to operate,cheap instruments,high sensitivity,good selectivity,and green environmental protection,it is gotten wide attention of the researchers.In this paper,on the basis of the resonance Rayleigh scattering method,using surfactants as sensitization and stabilization,dye molecules as probes,resonance Rayleigh scattering methods were established for the determination of chitosan(CTS).Amaranth(AMR),Congo red(CR),and Erythrosine B(Ery B)were selected as anionic dye,and sodium dodecyl sulfate(SDS),OP emulsifier and polyving akohol(PVA)were selected as sensitization.In the certain range of chitosan,the increased scattering intensities were linearly proportional to the increasing chitosan concentration,and they showed a good linear relationship.The experimental conditions were optimized,the sensitivity and selectivity of the method were discussed,and the mechanism of increasing sensitivity and stability of surfactant was discussed.Furthermore,the effect of the molecular weight and deacetylation degree of CTS on its accurate quantification was studied.The three methods were used for the determination of chitosan content in the same health product,and the results were consistent.On the basis of the resonance Rayleigh scattering method,using surfactants as sensitization and stabilization,dye molecules as probes,resonance Rayleigh scattering methods were established for the determination of Anionic surfactant(AS).Safranine T(ST)was used as cationic dyes,and polyvinyl alcohol(PVA)was used as sensitization.Under experiment conditions,the scattering intensities(?I)were proportional to the concentration of sodium dodecyl benzene sulfonate(SDBS)over the range of 0.15 ~ 5.00 ?g/m L,and the detection limit was 8.6 ng/m L.The optimum conditions of the RRS method and the mechanism of RRS enhancement were investigated.Accordingly,the method was applied to detect the contents of anionic surfactants in environmental water samples.The water samples were also detected by the national standard method,and there was no statistical difference in the results,with satisfied.Therefore,three resonance Rayleigh scattering method for the determination of chitosan and an resonance Rayleigh scattering method for the determination of anionic surfactant were established,with good sensitivity,accuracy and stability.They were applied to the determination of actual sample of chitosan and anionic surfactant,with satisfied results.
