Hericium erinaceus is a kind of famous fungus used as both food and medicine.Researches have shown that polysaccharide,which is the fermentation product of Hericium erinaceus,has anti-tumor,anti-oxidation,anti-aging,hypolipidemic and immunomodulatory functions,and it is used for the treatment of neurasthenia,gastritis and gastric ulcer clinically.At present,researches on relationship between the chemical composition and the physiological activity of fermentation products of Hericium erinaceus focus on the relationship between polysaccharide of Hericium erinaceus and hypoglycemic,and anti-tumor physiological activities.However,the antioxidant activity of polysaccharide of Hericium erinaceus was rarely reported.In this research,the related research group has been working on the systematic research of large-scale fungal-modified maize gliadin for many years,and the preliminary research results have shown that the fermentation products of medium,whose main component is maize gliadin,of Hericium erinaceus have certain physiological activity.Therefore,based on the previous research results,we work to research on the extraction and purification process of polysaccharide taking the fermentation products of Hericium erinaceus as raw materials,so as to clarify the antioxidant activity of polysaccharide of Hericium erinaceus through antioxidant tests.The main contents and results of tests are as follows:1. Optimization of liquid fermentation and solid-state fermentation process of Hericium erinaceus.Through screening He-02 liquid-cultured carbon source,nitrogen source,growth factor and culture process,as well as orthogonal tests and analysis,the optimum medium components of He-02 liquid culture are as follows:maize protein powder 12%,glucose 4.5%,potassium dihydrogen phosphate 0.35%,and vitamin B1 0.0015%.The optimum culture conditions are as follows:inoculation amount 8%,culture temperature 25°C,initial p H value 5.5,speed 150r/min.Tests have shown that with optimum medium components and under optimum culture conditions,the yield of polysaccharide are 29.37mg/m L,and the mycelial biomass is3.08g/100m L.Through screening He-02 solid-cultured carbon source,nitrogen source,growth factor and culture process,as well as orthogonal tests and analysis,the optimum medium components of He-02 solid culture are as follows:maize protein powder 30g,glucose 4.5%,Ca CO3 0.45%,KH2PO4 0.15%,and solid-liquid ratio 1:1.25.The optimum culture conditions are as follows:inoculation amount 16%,culture temperature 25°C,initial p H value 5.5,15days.2.Optimization of extraction process of mycelial polysaccharide and extracellular polysaccharide.Ultrasonic-assisted extraction was used to optimize the extraction process of mycelial polysaccharide from Hericium erinaceus.The optimum extraction conditions are as follows:ultrasonic power 600w,ultrasonic time 30min,ultrasonic temperature 80°C,ratio of raw materials to liquid 1:35,extraction times 3,polysaccharide yield 7.03%,an increase of 61.61%compared with traditional hot water extraction method.Orthogonal tests optimized ethanol precipitation conditions of fermentation broth of Hericium erinaceus,and the optimum extraction conditions are as follows:final concentration of ethanol 80%,p H value 6,athanol precipitation at 4°C for 12h,and polysaccharide extraction amount 17.35mg/m L.Ultrasonic-assisted extraction was used to optimize the extraction process of polysaccharide from solid-state fermentation products of Hericium erinaceus.The optimum extraction conditions are as follows:ultrasonic power 600w,ultrasonic time 30min,ultrasonic temperature 80°C,ratio of raw materials to liquid 1:30,ultrasonic extraction times 3,and polysaccharide yield 5.84%.3.Research on antioxidant activity of polysaccharide from Hericium erinaceus.Through determinations of reducing capacity,DPPH·free radical scavenging rate,O2-·scavenging rate,and·OH scavenging rate,the in-vitro antioxidant test results have shown that the fermentation broth polysaccharide of Hericium erinaceus has a strong ability of scavenging the free radical.Among the three kinds of polysaccharides in this experiment,the antioxidant capacity of He P3 is the highest.The results obtained in this research have provided basic data for the research of the function of fermentation products of Hericium erinaceus,technical basis for further clarifying the functional activity of fermentation products of Hericium erinaceus,and theoretical basis for the development of functional food of Hericium erinaceus.
