In this paper,Sargassum fusiforme polysaccharide was optimized by microwave-assisted water response,which were separated by ultrafiltration membrane technology.The antioxidant activity of SFPS(Sargassum fusiforme polysaccharide)in different molecular weight sections were analyzed.On the basis of this,in vivo anti-fatigue and mouse water maze experiment were analyzed,and the best group was chosed to be structural analysis.SFPS was extracted as raw material and microwave-assisted extraction technique.Based on single factor experiment,the optimum reaction conditions were extracted by response surface design method.The optimal conditions for the extraction of SFPS were as follows:liquid feed ratio 34 mL/g,microwave time 8min,water extraction temperature 95 ℃,extraction time 2.1h.Under this optimized condition,the yield of sfps was 11.51% ± 0.12%.SFPS were separated by ultrafiltration membrane(100,000、50,000、10,000、5,000)technology,and the antioxidant activity of SFPS in different molecular weight sections were analyzed.The results showed that the SFPS had different antioxidant ability in different molecular weight segments,and had some scavenging effects on hydroxyl radical,DPPH ·and superoxide anion radicals.SFPS on hydroxyl free radicals,DPPH · free radical scavenging effect is more obvious than superoxide anion.Free radical scavenging rate increased with the increase of polysaccharide concentration,showing a dose-effect relationship,and the smaller the relative molecular weight,the stronger the scavenging ability of free radicals.SFPSV,SFPSIV,crude polysaccharide were used to study the ability of learning and memory in mice,and 120 Kunming mice(male and female)were randomly divided into 12groups: normal control group,racemic anisodamine-induced memory impairment model group,piracetam(Naofukang)positive control group,experimental group,the experimental group were different molecular weight section of Sargassum fusiforme polysaccharide high dose group(0.4 g/kg),middle dose group(0.3 g/kg)and low dose group(0.2 g/kg).Continuous infusion of 15 d,Morris water maze test was performed to test the learning and memory ability of each group.20 minutes before the test,except the normal control group which intraperitoneal injection of saline,the other groups were injected racemic anisodamine,for which establishing a racemic anisodamine memory acquired disorder model,after the test,the serum T-AOC,SOD activity and MDA content were detected.The results showed that SFPSⅤ had a certain effect on the learning and memory ability of mice with memory impairment.Compared with the model group,the average escape latency was significantly shortened and the number of passes was significantly increased(P <0.05).The effect of polysaccharides from different molecular weight segments on the learning and memory ability of mice was different,and the effect of low molecular weight polysaccharides was the best(P<0.05).SFPSⅤ improved the learning and memory ability of racemic anisodamine memory acquired disorder model,the mechanism may be related to the antioxidant capacity of polysaccharides of Sargassum fusiforme;The utilization and activity of SFPS has a certain role.The anti-fatigue effects of SFPSV,SFPS Ⅳ,crude SFPS were studied in different molecular weight groups.The mice were randomly divided into 10 groups: normal control group,high molecular weight group(0.3 g/kg),medium dose group(0.2 g/kg),low dose group(0.1 g/kg).After continuous administration for 15 days,the activities of SOD,MDA,hepatic glycogen,muscle lactic acid and serum urea were recorded in mice with weight bearing swimming test.The results showed that SFPS had anti-fatigue effect,and the anti-fatigue activity of SFPS was the low molecular weight polysaccharide,which laid a foundation for further study on the mechanism of polysaccharide activity of Sargassum fusiforme,food development and utilization in the provision of new ways.SFPSV was eluted by anion exchange column chromatography with different concentrations of NaCl gradient to obtain three single polysaccharide components 0.1M SFPSV,0.3M SFPSV and 0.5M SFPSV,which were dialyzed and desalted by concentration.Polyglycol G-100 column was further eluted to obtain three charge properties,and molecular weight distribution homogeneous polysaccharide pure 0.1M SFPSVG-2,0.3M SFPSVG-1,0.5M SFPSVG-2.Through the infrared spectroscopy,three pure products have polysaccharide characteristic absorption peak,glycine hydroxyl stretching vibration peak,-CH3、-CH2 stretching vibration peak,polysaccharide hydrate vibration peak,glycosidic bond(C-O-C)asymmetric vibrational peak(3,6-ether bridge),S=O stretching vibration peaks(indicating thepresence of sulfuric acid substitutions).By ion chromatography analysis,the results show that0.1M SFPSVG-2 mainly in the presence of glucose components,0.3M SFPSVG-1,0.5MSFPSVG-2,there are five kinds of monosaccharides,namely glucose,fucose,galactose,xylose,mannose,the former two groups of peaks,the latter appeared a group of miscellaneous peaks.There were five kinds of monosaccharides in SFPS V by one-dimensional NMR spectroscopy.Among them,there were three kinds of α-type pyranose,two kinds of β-pyranosein in 0.1M SFPSVG-2.There are one α-type pyranose and one β-pyranose in 0.3M SFPSVG-1,0.5M SFPSVG-2.
