Hippophae rhamnoides is a dual-use plant for medicine and food.Xinjiang is one of the main producing areas of seabuckthorn in China.The annual output of seabuckthorn in Buljin County of Altay can reach 5 million tons.With the annual increase of cultivation of seabuckthorn in Xinjiang,it is particularly important to vigorously develop series products of seabuckthorn to promote rural economic development.In this paper,seabuckthorn was used as raw material to study the anti-oxidation,anti-aging,anti-fatigue and immune activity of seabuckthorn polysaccharides,in order to lay a foundation for the industrial development of seabuckthorn.The main results are as follows:(1)After extracting and optimizing the extraction of crude polysaccharides from Hippophae rhamnoides by ultrasonic extraction and ultrasonic assisted extraction,ultrasonic extraction-assisted extraction method was selected to extract seabuckthorn polysaccharides.The optimized process was:ratio of material to liquid 1:20(g/mL),temperature 73?,extraction time 22 min,intermediate frequency(48.1 kHz),extracted 2 times.Under this parameter,the extraction rate of seabuckthorn polysaccharides can reach 8.866%.The comprehensive indicators of hydrochloric acid method,by TCA method and sevag method were 62.22%,62.32%and 68.385%,respectively.(2)Hippophae rhamnoides polysaccharides have certain scavenging ability to DPPH free radicals,OHˇfree radicals and O2-ˇfree radicals,and their IC50 values were 1.55,0.97,7.95 mg/mL,respectively.When the concentration of seabuckthorn polysaccharides was the same as that of Vc(2 mg/mL),the reducing capacity of seabuckthorn polysaccharides was slightly higher.Seabuckthorn polysaccharide inhibited spontaneous lipid peroxidation and liver lipid peroxidation induced by CCl4,H2O2 and Fe2+-VC in mice liver homogenate,with IC50 values of 1.10,1.59,9.13,1.39 mg/mL,respectively.The results showed that seabuckthorn polysaccharide has good anti-oxidation effects.(3)At the same time of D-gal modeling,each group showed hair loss,slow movement,lethargy,hair color and yellow,the phenomenon of fighting in the cage was significantly reduced,and the performance of the model group was the most obvious.Compared with the model group,the low,medium and high dose groups of the seabuckthorn polysaccharide group can increase the index of heart,liver,spleen,kidney and brain,increase the activity of SOD and GSH-Px in vivo,and decrease the end products of oxidation.The pathological sections of the senile mice of the seabuckthorn polysaccharide group were observed by light microscopy,compared with the model group,the morphology of liver cells and the abnormality of liver tissue structure were improved.(4)The mice were swam with weight-bearing,referring to the blank group.The low,medium and high dose groups of seabuckthorn can prolong the weight-bearing swimming time by 29.87%,56.21%,and 37.64%,respectively.The content increased by 12.41%,30.75%,15.35%;BUN content decreased by 22.07%,25.89%,23.72%;LA content decreased by 4.40%,21.90%,22.47%.The results showed that seabuckthorn polysaccharide has good anti-fatigue effect.(5)Compared with the model group,the immune function evaluation test showed that the low,medium and high dose groups of the seabuckthorn polysaccharide group can increase the spleen index by 38.17%,96.19%,and 1.32 times,respectively.The thymus coefficient increased by 10.77%,33.11%,and 33.90%;the phagocytic index increased by 4.25%,16.05%,and 23.69%;the lymphocyte proliferation ability increased by 2.41,4.53,and 4.09 times;the hemolysin level increased by 0.41,2.09,and 2.00 times.The results showed that seabuckthorn polysaccharide has good enhancing immune function effect.
