Maca is a medicinal and edible plants,native to South America,Peru where the local Inca people used to cure diseases,and call it Peruvian ginseng.A lot of research work shows that the Maca polysaccharides have different physiological activities,such as anti-tumor,anti-oxidation,improve immunity,etc.,and with a huge development and utilization value.This paper optimized the method of Maca polysaccharides and the extraction process of Maca Polysaccharides.We also studied the basic physical and chemical properties of the polysaccharides and purified the Maca polysaccharides.The structure was studied by infrared spectroscopy.At last,we studied the acute toxicity of the powder and the polysaccharides.The main conclusions of this paper are as follows:1.Determined the extraction,isolation and purification of Maca polysaccharides.Optimized the conditions of hot water extraction.The main factors affecting the extraction rate of polysaccharides were studied by single factor test.Then,we use orthogonal experiment to optimize the extraction process and obtained that the solid to liquid ratio of 1:30under the conditions of 100?,2h extraction time in water bath extraction,polysaccharides yield can reach 8.9%.Through the enzymatic addition to starch,sevage method in addition to protein,dialysis and activated carbon decolorization for purification the Maca polysaccharides.2.The average molecular weight(Mn)of the homogeneous component in the range of relative molecular mass was 4.665×105.The IR spectra shows the characteristic absorption peaks of polysaccharides,and have the characteristic absorption peaks of pyranose,indicating that the constituent monosaccharides are pyranose.High performance liquid chromatography(HPLC)method to determine the maca monosaccharide composition of polysaccharides with glucose.3.Determined the method of content of Maca polysaccharides and the corresponding physical and chemical research.The content of the polysaccharides was determined by ultraviolet spectrophotometry(phenol-sulfuric acid method).we use glucose as standard materials and calculated the conversion factor with refined the Maca polysaccharides.The conversion factor is 1.75.Through experiments we find 1ml 5%phenol with 5ml sulfuric acid and color 30 min can get a better color results.At the same time,we verified the stability,reproducibility and precision of the phenol sulfuric acid method for the determination of Maca polysaccharides.The method was reliable.At the same time,we studied the appearance of maca polysaccharides,solubility,melting point,and identify the physical and chemical indexes.4.The acute toxicity studies were carried out on mice.The results showed that the lethal dose LD50>5.00g/kg.bw which was the actual non-toxic substance.The maximal tolerable dose(MTD)was performed at the maximum acceptable volume of the mice.The cumulative dose of the mice was 15g/(kg·bw)in 24 h,and no symptoms of poisoning were observed in the animals.There were no significant difference in the weight and organ/body ratio between the test group and the control group.There were no obviouspathological damage caused by the normal control group and the high dose group.It can be seen,the safety of the commercialization powder is very high,there were no obvious side effects,and there were also no obvious side effects of Maca polysaccharides.
