Objective: To investigate the novel non platinum metal prodrugs with biological activity,and to investigate their antitumor activity in vitro and the mode of action with DNA.Methods: Using different aromatic aldehydes to react with primary amine compounds and RuCl3,RuNOCl3 to get novel Schiff base Ru???complexs by direct synthesis,and their structures were characterized by nuclear magnetic resonance,infrared spectroscopy,mass spectrometry,thermogravimetric analysis,single crystal diffraction,and molar conductivity analysis.MTT method was used to study their antitumor activity in vitro and to investigate the interaction between target complexs and DNA.Result: Four ligands La,Lb,Lc,Ld and one novel bis Schiff base ligands Le containing nitrogen functional groups were developed,and Schiff base ligands of Le containing nitrogen functional groups were screened antitumor activities in vitro.Eighteen Schiff base Ru???complexes of 18,including four NO-Ru donors of 58 were obtained and that complexes 1,3,4 and NO donor 5,6,8 were screened to be sensitive to A549,HepG2 cells.Conclusion:The novel Schiff-base NO donor containing nitrogen functional groups of 10 showed superior anticancer activity,and other Ru???complexes of 1,3,4 and three Ru???-NO donors of 5,6,8 also showed preferable antitumor activity.The interaction between complex 10 and DNA is electrostatic.
