Schiff bases play an important role in biological systems with several applications.They represent an important class of compounds in medicinal and pharmaceutical field,showing biological applications including antibacterial,antifungal and antitumor activity.As indole scaffolds and Schiff base have exhibited favorable response as anticancer agents,these pharmacophores were subjected to structural modifications,to synthesize hybrid molecules and to investigate their anticancer potency which has been a core interesting strategy for the researchers.Many derivatives of indole-2-ones were also reported to possess good antimicrobial activity.N-1 and C-3-substituted indole derivatives have been found to play an important role in many biologically active compounds especially with anticancer activity.In view of these research results and because of the principle of active superposition,we synthesized four series of Schiff bases?BDH,BMH,DPH and BEH series?derived from 3-formylindole,3-acetylindole and N-methyl-3-formylindole.To our knowledge,the crystal structures of ten new compounds have not been reported.The products were characterized by melting point,X-ray,IR,1H NMR,13C NMR,UV-Vis.The fluroescence spectra of new compounds have been determined and exhibit favourable fluorescence properties.The in-vitro antitumor activities of synthesized compounds have been studied and their structure-activity relationships have been discussed.Meanwhile,the compound BEHFI was docked into DHODH?dihydroorotate dehydrogenase inhibitor?,and the interactions with the active site residues were analyzed.High Consensus Score indicates that BEHFI may have similar DHODH inhibitory activity and then similar pharmacological properties to Brequinar sodium,a famous DHODH inhibitor that is potent against many types of cancer.
