Objective:propolis is a widely used natural product produced by bees.It has many advantages,such as diminishing inflammation,killing bacteria and inhibiting tumor,etc and contributes greatly to the health of human beings.In recent years,cancer has become the most important disease of harm to human health,however chemotherapy is widely used as one of the effective methods for the treatment of cancer.Radiotherapy is currently the main way of cancer treatment,but the treatment side effects brought great harm to kill cancer cells as well as a part of normal cells.Propolis as a natural product with antitumor biological activity,under the condition of particular concentration,can induce apoptosis of breast cancer cells but very little toxicity to normal cells.This paper is designed to explore the optimization scheme of the ethanol extract of propolis flavonoids through orthogonal test;to observe the effects of ethanol extract from propolis of northeast black bees on the immune activity and the inhibition rate of tumor in Kunming mice and to investigate the antitumor activity of EEP.Methods: we prepared the H22 model mice,and the extraction of propolis from northeast black bees was carried out by single factor experiment and orthogonal test in Kunming mice.In the Carbon clearance test and anti tumor experiment,the mice were divided into five groups: negative control group,propolis high concentration group,propolis concentration group,propolis low concentration group and cyclophosphamide group.Continuous administration for 7 days,then they were weighed.The phagocytic index,thymus index and spleen index were measured in India ink.After the last administration,the rats were killed by cervical dissection,the organs were dissected and weighed,and the tumor inhibition rate was calculated.Results: the optimum method of ethanol extraction of Flavonoids from northeast black bees was that the temperature was room temperature,the solid to liquid ratio was 1:10,the ethanol concentration was 90% and the time was about 24 hours;Carbon clearance test in propolis dose group,the comparison of phagocytic index,thymus index and spleen index with negative control group had a statistical significance(p<0.05);propolis dose group tumor anti tumor experiment in weight compared with the negative control group,with a statistical significance(p< 0.01).Conclusion: the method of optimization method reduced the cost of extracting flavonoids,in line with the actual needs;carbon clearance test showed that the northeast black bee propolis extract could enhance the immune activity of H22 mice,anti tumor experiment showed northeast black bee propolis extract on the growth of tumor has a strong inhibitory effect to a certain extent.The experimental results showed that the ethanol extracts of propolis from northeast black bee propolis have significant anti-tumor effects.
