In recent years, cancer also known as malignant tumor has become one of major diseases that threaten human health.Due to the pollution of environment and increasing stress of work, the risk of cancer is gradually increasing. The traditional cancer therapies, including surgeon, chemotherapy and radiotherapy, have serious side effects. Therefore,a safe cancer therapy has to be developed.Photothermal therapy(PTT)that uses optical absorbing agent to kill cancer cells under light irradiation has reached a stage of clinic trials in recent years as a promising alternative or supplement to traditional cancer therapies.Compare to conventional approaches including radiofrequency or microwave ablation that were wildly used in clinic,PTT is significantly expected to improve the accuracy of treatment and reduce injury to normal tissue by using PTT agents as microscopic heating source. However, the most common cellular responses to this temperature is necrosis,a progress that compromises the integrity of cell membrane and releases intracellular constituents into the extracellular milieu producing detrimental inflammatory responses.Tremendous reports have achieved the co-therapy of inflammation and cancer, but few studies focus on inflammation caused by PTT,which becomes an urgent problem to be solved.Herein,an attractive strategy that AuNR-encapsulated graphitic(AuNR@G)nanocapsules combined with the prodrug P-aspirin was not only used to ablation the solid tumor but also to eliminate the inflammation caused by PTT was proposed. We synthesized AuNR@G nanocapsules by chemical vapor deposition method(CVD)and characterized its microstructure using variety of analytical instruments. We also synthesized prodrug P-aspirin,which combined aspirin and 1-Pyrene methanol through ester bond. The main contents are as follows:(1)We synthesized AuNR@G nanocapsules by CVD method, then characterized the structure of AuNR@G nanocapsules using variety of analytical instruments. We also used the Raman signal of the graphitic shell and two-photon fluorescence of AuNR@G for Raman imaging and TPL imaging,respectively.(2)We studied the NIR photothermal effects of AuNR@G nanocapsules. Then we synthesized the prodrug P-aspirin that combined aspirin and 1-Pyrene methanol through ester bond.P-aspirin was characterized by various analytical instruments.AuNR@G-P-aspirin complexes were developed by combining the AuNR@G nanocapsules and P-aspirin through ?-? interaction.(3)We further explored the in vivo NIR photothermal effects of AuNR@G and AuNR@G-P-aspirin complexes. Then, the in vitro and in vivo anti-inflammatory response of P-aspirin and AuNR@G-P-aspirin complexes was investigated, respectively.
