Vaccinium bracteatum Thunb.is a Chinese traditional medicinal plant which is also called Wufanshu.V.bracteatum leaves have many medicinal functions,including improving eyesight,hypoglycemic effect,anti-fatigue properties,anti-tumor properties and anti-aging properties.However,the main resources of this species are mainly wild resources because of the lack of artificial breeding.In this research,the V.bracteatum were grown under four shade treatments(0,25,50,and 75%of natural incident irradiance)from June to August in Hangzhou.The photosynthetic physiology,nutrient content and antioxidant activity of leaves are compared to prove which shade treatment is best for the growth of V.bracteatum.Response surface methodology(RSM)was used for optimized extraction for antioxidant activity of V.bracteatum leaves.Least squares vector support vector machine(LS-SVM)was used for nondestructive evaluation techniques to project the nutrient content of extracting solution of V.bracteatum leaves.And the chemical composition of extracting solution was identified based on Waters ACQUITY UPLC/SYNAPT G2-S HDMS.Finally,the nutrient content and antioxidant properties of Wu Mi Fan of different treatment were compared by in vitro digestion.(1)The protected cultivation research was carried out in Hangzhou from June to August.The results showed that V.bracteatum with 0%shading appeared leaf scorch.The nutrient content and antioxidant activity of leaves were higher than other treatment because of the oxidation defense mechanism.But the photosynthetic physiology was at low level.The V.bracteatum with 50%shading showed a high level of photosynthetic physiology,nutrient content and antioxidant activity.So 50%shade treatment is the best for the growth of V.bracteatum.(2)The results of extraction research using RSM showed that the optimum values of the antioxidant activity was X1(extraction temperature)=63.8 ~o C,X2(extraction time)=40.2 min,X3(liquid-solid ratio)=68(v/w).The yield of total phenolics,total flavonoids and anthocyanins were 29.25,139.50 and 5.39 mg g~-11 Dry Weight(DW),respectively,and the ascorbic acid equivalent of DPPH radical scavenging capacity was 26.54 mg Ascorbic acid g~-11 DW.The results of nondestructive evaluation techniques based on LS-SVM showed that DPPH free radical scavenging ability and anthocyanin content could be predicted by LS-SVM model based on RGB color space.(3)In this research,sorghum curcumin,apigenin,quercetin,citric acid,chrysin and theophylline were identified from extracting solution of V.bracteatum leaves.The results also showed that heating could improve the antioxidant activity and total flavonoids of the extracting solution.The total flavonoids,DPPH radical scavenging activity and ABTS radical scavenging activity of the treatment at 100 ~o C were increased by 56.27%,64.33%and 80.26%compared with CK group.And the color of Wu Mi Fan of the treatment at 100 ~oC was better.The results of in vitro digestion showed that the total flavonoids and ABTS radical scavenging activity of the treatment at 100 ~oC were increased by 43.40%and 32.54%,respectively,compared with CK group.They also were increased by 120.10%and 42.28%compared with sticky rice,respectively.Therefore,it was suggested that heating the extracting solution of V.bracteatum leaves that increase antioxidant activity and nutrient content.
