Osmund is Osmundaceae Osmundaceae perennial herbaceous fern,also known as Osmunda.Due to years of reproductive accumulation,now the product is rich in resources in our country,the production is enormous,actively in the export market.With the increasing domestic attention of osmund,studies have shown that the active component in the physiological function as antioxidant,free radical scavenging,anti-aging effect,even widely circulated on the food industry,is a kind of promising natural antioxidant materials.In this paper,we take osmund which has strong antioxidative activity as research object.In the study of osmund components as the basis,the antioxidant activity as index,using traditional extraction and extraction techniques to segment the flavonoids of osmunda japonica.And to study the antioxidant activity of each segment,so as to have a certain significance both on finding a new and efficient natural antioxidants and the development and utilization of osmund resources.Therefore,this paper around the active ingredients of osmund carried out the following aspects of research:Comprehensively studied the total amount of various components of osmund: used the rutin ethanol method to determine the total flavonoids in osmunda japonica was 3.4%,used the tungsten molybdophosphate casein method to determine the total tannin content was 3.17% in osmund,used phenol sulfuric acid method to determine the total polysaccharide content was 1.57% in osmund,used anthrone maltobiose colorimetric method to determine total glycoside content in osmund was 1.51%,and the experiments were carried out with precision,stability and repeatability,the results were in line with the requirements of the theory.The content of flavonoids in osmund as indicators,with ethanol concentration extraction process,the extraction temperature,extraction time,extraction agent concentration and liquid ratio for single factor,studied the optimum extraction scheme,and compared the effect of different factors on extraction rate,size,results showed that in 80% ethanol concentration,liquid ratio 1:25,extraction time 1.5h,extraction temperature 65?,under this condition the rate of flavonoid was the highest,is 15.933mg/g,the influencing degree of every factor was extraction time > temperature > solid-liquid ratio,ethanol concentration,consistent with theoretical predictions,explained that the process is suitable for extraction of flavonoid compounds in osmund.Using DPPH Spectropho-tometric Assay to analysis extraction of flavonoid compounds in osmund,the compounds were divided into four polar segments: petroleum ether,ethyl acetate,n-butyl alcohol and water,determined each DPPH free radical scavenging ability of polarity period,compared with Vc solution,the results showed that ethyl acetate fractions were significantly higher than those of other parts in scavenging free radicals.When the concentration was 1mg/ml,the clearance rates were 96.67%,respectively.Results proved that Osmunda japonica has strong antioxidative activity,and the active material had a large antioxidant activity in theethyl acetate section.The experimental results for provide a theoretical basis for taking Osmund as raw materials to develop the natural,efficient,non-toxic antioxidants.
