Wheat germ is an important part of wheat seed and the source of wheat growth and development.Wheat is the main food crop,wheat is unusually rich resources in our country,wheat germ as a by-product in the process of wheat processing,development and utilization of wheat germ has long exist industrialization,standardization,lack of the status,the use of wheat germ has not been effective.This topic from the wheat germ albumin extraction,purification,wheat germ albumin physical and chemical properties and characterization of functional properties of wheat germ protein production plant design study of wheat germ albumin,purpose is to increase the added value of wheat processing,in order to be able to reduce the high quality food waste of resources.In order to provide a basis for further research on wheat germ albumin,the main research contents include the following contents:(1)In this paper,through the extraction of wheat germ protein,water was studied by single factor and orthogonal analysis of wheat germ albumin in the process of extraction process control: wheat germ albumin was studied in the process of extracting liquid ratio,extraction temperature,extraction time,extraction solution pH's influence on the extraction yield of wheat germ albumin.Obtained by optimization experiments of various conditions optimum parameters of wheat germ albumin extraction as follows: the material liquid ratio and extraction time,extraction temperature 35 ? 90 min.Under this condition,the extraction rate of wheat germ albumin can reach 27.39%.The control of production process parameters during the process of wheat germ albumin can effectively improve the extraction rate of wheat germ albumin.(2)The experiment based on the analysis of wheat germ protein solubility,a water-based,oily,foamability,molecular size analysis,scanning electron microscope,secondary structure and thermal characteristics and the function of lead the wheat germ albumin were characterized.Wheat germ albumin with good solubility,whenthe pH is less than 4,as the pH increase solubility decreases gradually,wheat germ albumin solubility worst when pH is 4,when pH greater than 4,with the increase of pH,solubility increased.The wheat germ albumin has good water holding capacity,but the oil is poor,the wheat germ albumin has good foaming property,but the foam stability is poor.Gel electrophoresis showed that wheat germ albumin has 14 bands,molecular weight of 97 Ku respectively,82 Ku,70 Ku 51 Ku,47,65 Ku,Ku,40 Ku,35 Ku,30 Ku,26 Ku,18 Ku,13 Ku,9 Ku,4 Ku.The infrared spectra were used to determine the protein content of the wheat germinal protein with 21.28% of the beta-folding,13.61% of the random crimp,15.74% of the a-helix,33.15% of the beta-Angle and 16.12% of the reverse eversebeta-folding.The DSC method was used to determine the denaturation temperature of wheat germ albumin at 55.59?,with good thermodynamic stability.Wheat germ albumin has the ability to absorb lead ions,and each 1mg of wheat germ protein can absorb 0.0024 mg of lead ion effectively.The precipitation quality of wheat germ albumin was determined by different precipitation methods,and the precipitation effect of ethanol precipitation was found to be the best.(3)It was identified by high precision mass spectrometry to contain 1045 protein groups containing 2224 proteins.The GO functional annotation of wheat germ albumin showed that the wheat germ albumin involved 12 molecular functions,15 kinds of cell composition and 18 biological processes.Wheat germ albumin mainly involves the DNA binding,and catalytic activity,structural molecule activity such as molecular function,cell organelle,cell functions such as be,metabolic process,the cellular process of biological processes,etc.KEGG Pathway analysis was performed on the protein identified,and it was found that wheat germ albumin was involved in216 pathways.The main metabolic pathways involved in wheat germ protein were Ribosome,Carbon metabolism,Biosynthesis of amino acids,Glycolysis/Gluconeogenesis,Spliceosome pathway.(3)The production design of wheat germ albumin,complete productiontechnology of wheat germ albumin and workshop design,to become in the process of production of each process has a specific description,selection of equipment for production,and made the process flow diagram,equipment layout,workshop section,the factory layout and main equipment drawing.The material balance and energy balance in the process are calculated in detail.Develop the product enterprise standard,the final production plant design can be put into production.Through the experiment of extracting conditions and properties of wheat germ albumin for further understanding,complete the production design of wheat germ albumin,for the development and utilization of wheat germ albumin and wheat processing to provide a certain basis.
