In this project,sesame,peanut,rapeseed and camellia seed are used as raw materials,and the oils are prepared by hot pressing,cold pressing and refining.The basic physicochemical indexes,fatty acid composition analysis,trace tocopherols,sterolsand polyphenols content of the oils obtained from different processes were preliminary analyzed,the effects of sesame oil with different technologies on the anti-aging ability of 2BS cells and the effects of different technologies of sesame oil on blood lipid reduction in SD rats were studied.The main results of this study are as follows:(1)After high-temperature roasting of the oil,the obtained oil has darker color,lower moisture content,higher acid value and peroxide value,lower tocopherol content and sterol content,and higher total phenolic content.After the refining process of sesame oil,the free fatty acids in oils and fats are effectively removed and the antioxidant substances in the oil are decomposed and reduced,so the acid value and peroxide value of the refined sesame oil are reduced compared to the cold-pressed sesame oil;In addition to adsorptive pigments and colloidal impurities,activated clay has a strong adsorption capacity for polar substances,so a certain amount of tocopherols and sterols will be lost during the adsorption and decolorization stage.The loss rate of tocopherol in the process of adsorption decolorization and high-temperature deodorization is the highest,and the loss rate of sterols in the alkaline deacidification and adsorption decolorization stages is the highest,so the degree of refining of the oil should be reduced.(2)In the study of sesame oil delaying the aging ability of 2BS cells,it showed that: when the concentration of sesame oil extract was 10 mg/m L-90 mg/mL,the viability of 2BS cells increased with increasing concentration of sesame oil extract,and the vitality of 2BS cells treated with cold-pressed sesame oil extract was the highest,indicating that compared to the hot pressed sesame oil extract and refined sesame oil extract,cold pressed sesame oil extract can promoter cell growth and proliferation better.The content of MDA in the blank group was 0.68 nmol/L.After addition of sesame oil extracts to the 2BS cells,the MDA content decreased,and when the sesame oil extract was added at the same concentration,the cold pressed sesame oil extract hadthe best reduction on MDA content.Theresults show that adding sesame oil extract can reduce the extent of cell damage,and the effect of cold-pressed sesame oil extract is the best.The T-AOC level of 2BS cells in the blank group was 0.24 U/m L.When the cold-pressed sesame oil extract and the hot-pressed sesame oil extract were added,the T-AOC level of the 2BS cells increased slightly,but the concentration of sesame oil had no obvious regularity.When the refined sesame oil extract was added,the T-AOC level and the blank group of the 2BS cells did not change much.When the concentration of sesame oil was 60 mg/m L-90 mg/mL,the T-AOC level of 2BS cells was the best when adding cold pressed sesame oil extract.It can be concluded that sesame oil of different technologies contains antioxidants,and the total antioxidant content in cold-pressed sesame oil is higher than that of hot-pressed sesame oil and higher than refined sesame oil;therefore,the sesame oil of different technologies delays the aging of 2BS cells.The cold-pressed sesame oil extract has the strongest anti-aging ability.(3)Within a certain dose range,cold-pressed sesame oil can reduce the serum TC level in high-fat rats,but the effect is not significant(P>0.05),and it can significantly reduce the serum TG content in high-fat rats(P<0.05),and can significantly reduce the high serum AST levels(P<0.05)and significantly reduce ALP levels in serum of rats with high-fat(P<0.05);significantly decreased MDA levels in the liver of rats with high-fat(P<0.05),significantly improved rats T-SOD level and T-AOC level in the liver(P<0.05),and the effect is better than hot-pressed sesame oil,better than refined sesame oil,it is a kind of fat and oil that is beneficial to human health in the experimental study of blood lipid reduction in SD rats.Pathological sections of SD rats showed that the livers of the basic group SD rats were reddish-brown in appearance,and the liver tissues were smooth and elastic.No fat vacuoles were observed.Under the microscope,the structures were normal and without abnormalities.In the high-fat group,the color of the liver was darker than that of the basic group,the volume of the liver was significantly larger,and a lot of fat particles were attached to the liver.Under the microscope,it can be seen that there are different degrees of steatosis in the rat liver,and there are different kinds and different sizes of fat vacuoles in the cells,accompanied by cell fusion.Compared with the high-fat group,the hepatic steatosis of the SD rats in the experimental group was alleviated,and the fat vacuoles in the hepatocyte structure gap were reduced,and the effect of the cold-pressed sesame oil group was better than that of the hot-pressed sesame oil group and the refined sesame oil.It shows that gavage the same dose of different technology sesame oil,crushed cold sesame oil for SD rats blood lipid reduction effect is more significant.
