In our country,a large amount of collagen in food raw materials has not been used effectively.For more effective use of collagen in food raw materials and added value,it is significant to prepare collagen membrane with collagen in food.The methods of preparation of collagen membrane mainly include drying methods and freeze-drying methods.However,there are some disadvantages such as uneven thickness,uncontrollable microstructure,low mechanical strength,and uncontrollable degradation time,which limited the collagen's application.Electrostatic spinning technology is a new processing method for preparing nanoscale ultrafine fiber based on polymer solution spraying under electric field.But the product is short in low mechanical strength and short degradation time.Therefore,it is urgent to establish a spinning method to improve mechanical strength,controllability of degradation time and homogeneity of spinning diameter.Collagen(Co L)was gained from the bovine tendon,and collagen(CoL)membrane was prepared using electrospinning technique with hexafluoroisopropanol as the solvent.The sync hronous crosslinking process was carried out during electrospinning with glutaraldehyde(G A)as crosslinking reagent.And then optimize the collagen concentration,GA dosage and spi nning condition.And evaluate the physicochemical and biological properties of the membrane by various characterization methods.Collagen was dissolved by hexafluoroisopropanol solution in percentages of 4%,5.5% a nd 7% and spray spinning with GA solution in percentages of 4.5%,9% and 18%.And then c haracterize the physical and chemical properties of the spinning by different means.For exam ple: observing the spinning diameter and spinning density by SEM,detecting the effect of G A crosslinking on collagen structure by FTIR,analyzing the influence of different GA concen tration on the crystal configuration of collagen,evaluating the influence of GA dosage on the mechanical properties of collagen by texture analyzer,evaluating the effect of GA dosage on hydrophilicity of collagen spinning material by testing the contact Angle of the scaffold mater ial.In addition,the biological properties of the spinning film were characterized in cytotoxicit y,biocompatibility and cell proliferation.And the security of the collagen membrane in food packaging,water soluble and water vapor evaporation rate were tested.The results indicated that optimal effect of spinning exists when concentration of collage n was 5.5%,the voltage was 20 kV,the flow rate was 2mL/h,and the receiving distance was 12.5cm.The diameter and spinning density of collagen spinning increased with the increasing of GA content,and when the concentration of the GA was 18%,we get the largest diameter w ith the collagen concentration remaining,which was about 500~800 nm.SEM result indicated that the diameter of fiber was most uniform when the GA concentration came to 9%.FTIR re sult indicated that The triple helix structure characteristic of collagen has not been destroyed d uring the spinning process.But the XRD result indicated that the crystallinity of collagen redu ced after crosslinking.The testament of mechanical performance indicated that the spinning el asticity first increases and then decreases with the increasing of GA.The spinning membrane contact angle indicated that the contact angle became larger as the GA concentration increase d,which means the hydrophilicity of spinning was increased,which related to GA's hydropho bicity.The results of biological characterization indicated that the cytotoxicity of the spinning film was 0,and there was no significantly difference between the experimental group and the control group in proliferation(P > 0.05).Detection in food packaging applications indicate t hat the material conforms to the safety of the national standard,with low water solubility and water vapor evaporation rate.In the research,a kind of collagen fiber with suitable density and diameter was obtained by electrospinning blending solution of Co L and GA,which optimized the process of collagen spray spinning and provided references for the application of collagen membrane in food and biological materials.
