As a simple and safe new technology for preparing biomaterials,electrospinning technology can not only prepare nano-sized fiber materials,but also affect the morphology and surface properties of fibers by controlling parameters.This research topic took tilapia collagen(FC)and polycaprolactone(PCL)as basic materials,and mainly focused on the "preparation and Performance Research of fish collagen based electrospinning membrane".(1)The preparation process of FC-PCL nanofiber membrane mainly included the effects of solution concentration,spinning aid PEO content,dissolution time,spinning solution properties and spinning time on the spinning process and fiber morphology.Solute content,PEO content,FC/ PCL ratio and spinning time all affect the spinning process and fiber morphology by changing the properties of electrospinning solution,that is,the viscosity and conductivity of the solution.For this system,the effect of solution viscosity on its spinnability was much greater than that of conductivity.At the same time,the electrospun solution parameters that can form good fibers in this system are determined,that is,when the solution viscosity range was about 80?140 m Pa·S and the viscosity was about 120 m Pa·S,the diameter of the material fiber prepared under this condition reaches the nanometer level.(2)FC-PCL nanofiber membrane was prepared and characterized with PEO as sacrificial fiber,mainly included the determination of swelling rate,hydrolytic stability,FTIR,contact angle,tensile breaking strength and porosity.The results showed that the removal of sacrificial fiber would reduce the swelling rate and tensile breaking strength of the material,and improve the hydrolytic stability,hydrophilicity and porosity of the material.FTIR results showed that PEO sacrificial fibers was successfully removed by cleaning.(3)The biosafety test of materials mainly included cytotoxicity test,live/dead cell staining,cell material co culture,hemolysis test and subcutaneous implantation to detect the degradation and safety of materials in vivo.The results showed that the material had no cytotoxicity and hemolysis,and could promote cell adhesion and proliferation,and support cell infiltration and tissue vascularization after implantation in vivo.The results showed that the macroporous material prepared with PEO as spinning aid and sacrificial fiber overcomes the disadvantage of dense accumulation of fibers prepared by traditional electrospinning method.It is a kind of tissue engineering biomaterial with good biocompatibility...
