This experimen used carbonization method to synthesize Ganoderma lucidum hollow microspheres.Then,the magnetic microspheres were prepared by using Ganoderma lucidum hollow microspheres loaded magnetic nanoparticles.With scanning electron microscopy?SEM?,transmission electron microscopy?TEM?,energy spectrum analysis?EDS?and Fu Liye transform infrared spectroscopy?FTIR?analysis of Ganoderma lucidum spore hollow microspheres,the experimental results show that after high temperature carbonization,the morphology is complete,there are cavities in the interior,and there are organic functional groups and chemical bonds on the surface.With transmission electron microscopy?TEM?,X-ray diffraction?XRD?,vibrating sample magnetometer?VSM?and nitrogen adsorption desorption?BET?and other modern means of science and technology of magnetic microspheres,showing that the magnetic Ganoderma lucidum spores has nanoparticles inside,its composed of Fe and O elements,it's Fe3O4;magnetic properties,with a remanence low?Mr=0.53 emu/g?and high coercivity?Hc=58.7 Oe?,can be quickly separated into magnetic field.The magnetic microspheres of ganoderma spores with H2O2 can catalyze the oxidation of peroxidase substrate TMB blue material,its absorption peak at 652 nm,this result proves that it has peroxidase activity,the optimum pH is 4,the optimum temperature is 30?-35 ?.Using TMB as the substrate,the Michaelis Menten kinetic reaction is sucessful and.Km=0.095 mM;magnetic ganoderma spore microspheres can also catalyze the decomposition of H2O2 into oxygen,which proves its catalase activity.Magnetic ganoderma spores can be used as adsorbents to remove difficult degradation of Congo red dye,the adsorption process accords with the quasi two level adsorption rate equation and Freundlich adsorption isotherm model;thermodynamic analysis indicates that adsorption of Congo red ganoderma spore magnetic microspheres is a spontaneous process.Magnetic ganoderma spores can be used as a carrier of immobilized CotA protein,the performance degradation of orange ? CotA protein catalyzed degradation;The 10 mg immobilized CotA protein can completely degradate orange?with the concentration of 50 mg/L in 60 min,and after recycling many times,the loss of enzyme activity is only 16%;CotA protein immobilized on the surface has good decolorization ability and reuse.
