By using wild Oregon-R Drosophila melanogaster as the experimental object, the effects of different concentration of spore oil from Ganoderma Lucidum (GLSO) and different ratios of sugar/protein on the lifespan in Drosophila were implemented, respectively.1. The effect of GLSO on Iifspan in DrosophilaThe lifespan change of Drosophila fed base diet and test diets addedd with GLSO1.25g/L (H),0.625g/L (M),0.3125g/L (L) were investigated. Fresh medium was changed twicely a week, meanwhile, the dead flies of different groups were recorded. Data was calculated with SPSS v19.0.By raising Drosophila fed tested diets as above with or without0.5%FD&C No.1blue, the effect of GLSO with different concentrations in Drosophila was conducted. Drosophila with same age emeged within8hours were set into vials with1female and1male for mating5days. After the new offerspring emering, the offsping amount of per vial with diffrenr diets was recorded in days. The reproductive capacity in Drosophila fed diffrent diets added GLSO was compared with the total emering ofspring flies per vial.21days old Drosophila collected from diffrent treatments and frozen under-80℃were used to measure activity of SOD and CAT, and MDA content with kits. The effects of different GLSO concentration on the activity of SOD, CAT and the MDA content in Drosophila were analyzed, respectively.It was showed that the mean lifespan of male Drosophila in different groups, H, M, L and base were44.23±0.689(d),44.47±0.69(d),42.85±0.61(d), and42.77±0.75(d) respectively. Compared with base dite, the mean lifespan of group H, M and L were extended3.43%,3.97%and0.19%, respectively. The mean lifespan of female Drosophila in different groups, H,M, L and base were45.31+0.65(d),45.22±0.78(d),43.01±0.82(d) and41.37±0.80(d), respectively. Compared with base dite, the mean lifespan of group H, M and L were extended9.54%,9.32%and3.97%, respectively. The results suggested that GLSO had benefit effect for extending lifespan of both genders in Drosophila. The better GLSO content is H and M, which were better for female. GLSO could affect the intake quality in Drosophila. Compared with male flies fed base diet,It was showed that the intake quality in Drosophila was affected by GLSO. Compared with base, the intake quality of H, M, M group in male flies were reduced by45.76%,53.39%and20.34%, respectively, and the intake quality of H, M, M group in female flies were reduced by19.57%,19.47%and28.62%, respectively. GLSO could increase the reproductivive capacity in female flies. Compared with base, the reproductivive capacity of H, M, M group in female were increased by19.57%,19.47%and28.62%, respectively. High level (H) GLSO could improve the activities of total SOD and Cu Zn-SOD, and reduce MDA content in Drosophila. GLSO could improve CAT activity in Drosophila as well.2. The nutritional interfering effect of different ratio of dietary sugar/protein on the lifespan in Drosophila.Female flies were fed diets with different proportion (%) of sugar(S) and yeast (P) as bellows:2/2(S/P),2/4,2/8,2/16,4/2,4/4,8/2,8/8and16/2. The dead flies were recordedconsecutively fed test diets, which were analyzed the effect of the sugar/protein ratio on lifespan in Drosophila. Meanwhile, curcumin of1.0mg/Kg was added in those test diets as above, which were used to investigate the interfering effects of curcumin on Drosophila fed high calorie.It was showed that dietary sugar/protein ratio could significantly affected lifespan in female Drosophila. Under the condition with same sugar proportion in diet, the increasing of dietary protein intake could reduce mean lifespan in Drosophila. Under the condition with lower protein proportion in diet, the raising of dietary sugar intake could increase mean lifespan in Drosophila. Supplementing curcumin in diet with high sugar proportion could increse lifespane in Drosophila, of which the effect is better when the diets with sugar of4%and8%. However, Curcumin had not interfering effect in Drosophila fed diet with high protein proportion. This thesis firstly reported the anti-aging effect of GLSO on Drosophila, the effect of sugar/protein ratio on lifespan in Drosophila, the literfering effect of curcumin on Drosophila fed diets with high calorie.
