Immune Regulation Effects Of Seven Polysaccharides In Vaccinated Chickens With IBD Vaccine

Objectives:To discuss the immune regulation effects of seven polysaccharides in vaccinated chickens with IBD vaccine,which will provide the theoretical basis for the use of IBD vaccine,in the practical production.Methods:270healthy male broilers of1day-old were randomly divided into9groups (Ⅰ, Ⅱ, Ⅲ Ⅳ, Ⅴ, Ⅵ, Ⅶ,Ⅷ,Ⅸ) of30broilers each, group Ⅰwas designed as control, Group Ⅱ, was designed as vaccine control.When7days,in group Ⅲ, Ⅳ, Ⅴ, Ⅵ, Ⅶ,Ⅷ,Ⅸ, every broiler was treated by seven polysaccharides injection with1.0ml, respectively, for three successive weeks. From Ⅱ to Ⅸ group, the first and second immunity was respectively at12days and22days, for every broiler. At7、14、21、28,35and42days, chose5broilers of each group, weighed, then collected blood by heart puncture technique to obtain plasma and blood serum, collect spleen, thymus and the bursa of fabricius, then weighed. In the experiment, AGP antibody titer, immune organ index,the activity of macrophage cell, the density of B cell and T cell,the density of CD4+T cell and CD8+T cell and the ratio of CD4+/CD8+were detected,in each group.Results:1、At21,28and35days,the thymus index,the bursa of fabricius index and spleen index of polysaccharide injection group,was higher than that of control group(P<0.05),significantly; between astragalus polysaccharide injection group and other polysaccharide injection groups is significantly different (P<0.05), in addition to the astragalus polysaccharide injection group, the other6kinds of polysaccharide injection group, no significant difference.2、At14,21,28and35days, there is no significant difference in the control group, the AGP antibody level of the polysaccharide injection groups are higher than the control group, the extremely significant difference with the control group (P<0.01), the differences between the astragalus polysaccharide and other polysaccharide injection groups are significantly (P<0.05), in addition to the astragalus polysaccharide group, there are no significant differences between other6polysaccharide injection groups. 3、At14,21and28days, the polysaccharide injection groups which used with ND vaccine, the activity of macrophage cell is significantly higher than the control group (P<0.05or P<0.01); After two hares, at28days, the index of macrophage cell of the polysaccharide injection groups significantly increased, significantly higher than14and21days (P<0.05), and there is not very significant difference between polysaccharide injection groups(P>0.05).4、At21and28days, the density of B cell and T cell of the polysaccharide injection groups is significantly higher than the control group (P<0.05).At7and14days, the peripheral blood T and B lymphocytes quantity of each polysaccharide injection group is more than the control group, but no significant difference (P>0.05).5、At14,21and28days, the CD4+T lymphocyte percentage of the polysaccharide injection groups was significantly higher than that of control group (P<0.05), on21-day-old, the percentage of CD4+T lymphocytes of the astragalus polysaccharide and epimedium polysaccharide group were significantly higher than that of control group; there is no significant differenc between the percentage of CD8+T lymphocytes of the polysaccharide injection groups and that of control group (P>0.05); the ratio of CD4+/CD8+of the polysaccharide injection groups was significantly higher than that of the control group (P<0.05).Conclusion:The study shows that the seven kinds of polysaccharides can significantly increase the immune function of boilers, at the same time, can markedly improve the immunity response of IBD vaccine in boilers when used with IBD vaccine. The effect of astragalus polysaccharides is most obvious.
