In recent years, Flammulina velutipes draws lots of attentions from people all over the China as a kind of healthy and organic food. The factory production developed rapidely in the background of Flammulina velutipes is popular among Chinese people. The strain of Flammulina velutipes as the most important part in the factory production, however is depended on importing from overseas. That suituation has cause big problems in China's Flammulina velutipes export because of the intellectual property right is not belonged to us. Therefore, the Flammulina velutipes breeding is vitual to us and breeding out Flammulina velutipes strains that we have the intellectual property right is very urgent.Flammulina velutipes has been divided into two kinds on the behalf of the color:the white and the yellow. Wild type of Flammulina velutipes'color is yellow and the white Flammulina velutipes is one of the mutants. The yellow strain has the advantage in growing speed, temperature, smell etc. The white strain has the advantage in even, mouthfeel, stabale etc.In this research, we collect 14 white Flammulina velutipes strains and 4 yellow Flammulina velutipes strains from different factories and places. Firstly, we selected 4 good white strains and 4 good yellow strains as parent strains through cultivation experiment, rivalry experiment, SRAP and ISSR DNA molecular marker experiment, mycelial activity test etc. These parent strains have the advantage in growing speed, economical character and so on. Secondly, we get 47 monokaryotic strains through protoplast monokaryotic 29 times. Then we use these 47 monikaryotic strains to do Mon-Mon hybridization and Di-Mon hybridization. In Mon-Mon hybridization, we made 1084 pairs combination and get 84 hybird strains. In Di-Mon hybridization, we made 104 pairs combination and get 19 hybird strains. We get 103 hybird strains in total. Thirdly, we select 70 hybird strains in those 103 strians through plating experiment, mycelial activity test etc. At last, we cultivate that 60 hybird strains and found hybird strains 3750-1,3750-2 and 3753-3 have advantage in product mass, color and economical charaters.
