In recent years,enterprises engaged in Chinese herbal drug production and processing are increasing with the revival and development of traditional Chinese medicine.However,the disposal of a large number of residue generated from the extraction of Chinese herbal drug was a difficult problem need to be solved.It maybe pollute the environment seriously if was not disposed properly.In addition,the residue still remains a lot of active ingredients and large amounts of nutrient substance.In the past,the main procedure for disposing the Chinese herbal drug residue were landfill and incineration,which may cause environment pollution,all of which has high value of resource reuse.Therefore,the study found that using microbial fermentation residue or production of feed additives.But the traditional fermentation process of fermentation temperature,fermentation time and pH value of the water content of substrate were quantified,resulting in poor fermentation process.The orthogonal experiment although the fermentation conditions were screened,but only orthogonal test optimization on the condition of single point,the lack of the study of various factors interaction,cannot find the best combination in the whole region.In order to improve the application effect of Chinese medicine residue as animal feed additives,the dregs of the active ingredients in the greatest degree of precipitation,this study using the response surface method was used to investigate the conditions of Astragalus residue mixed solid fermentation.In this study,the effect of fermentation on the precipitation of the main components of Astragalus residue was explored to improve the value that traditional Chinese medicine residue applied in feed additives of animal,the technological conditions of the mixed solid of Astragalus residue were explored.In this study,the Astragalus residue was used as raw material,Aspergillus niger,Bacillus subtilis with the advantage of cellulose degradation used for fermentation.The Astragalus residue was implemented with fermentation processing,the Nitrogen source type,nitrogen source added amount,Astragalus residue matrix particle size was screened by evaluating indexes including the enzyme activity,Carrying amount of bacterium quantity and the content of astragaloside.Optimal technology of mixed bacteria solid fermentation of Astragalus residue was acquired by single factor experiment was used to optimize the solid fermentation conditions including the proportion of mixed bacteria,the amount of inoculation,fermentation time and water content,and the optimum inoculationamount,fermentation time and water content were optimized by response surface designed with Box-Benhnken,and the response value was cellulase activity.The result of this study was provided below.(1)The optimum nitrogen source was urea,and the optimum adding amount was 0.3%,and the best particle size of Astragalus residue ranged from 5 to 10 mesh.(2)The result of single factor experiment indicated that the best ratio of Aspergillus niger to Bacillus subtilis was 1:3,the optimal inoculation was5%,and the best water content was 60%,and the time for fermentation was 6 days.(3)The optimum technological conditions for solid fermentation of Astragalus residue was acquired by response surface,the particle size of Astragalus residue ranged from 5 to 10 mesh,0.3% urea used for nitrogen source,ratio of Aspergillus niger to Bacillus subtilis was 1:3,inoculation amount was 5.41%,water content was 60%,and the time for fermentation was 6 days.(4)The solid fermentation of Astragalus residue was implemented with the best procedure,and the result indicated that the cellulase activity was 4493.21 U/g,the amylase activity was 75.12 U/g,and the acid protease was 113.52U/g,the amount of the precipitation(0.67±0.01)mg/g,the content of Bacillus subtilis was4.605×107 cfu/g,1.805×106 cfu/g of Aspergillus niger.The optimization process of Astragalus residue is a mixture of the solid fermentation and good growth of Aspergillus niger and Bacillus subtilis,cellulase production,effective degradation dregs cellulose,and promote the precipitation of astragaloside IV in the dregs,using this technology fermentation Astragalus medicine slag production of feed additive has practical significance.
