In this experiment,different doses of the purified ?-conglycinin(7S)and glycinin(11S)were added to the diets of weaned piglets in order to build sensitization model of piglets.ELISA method was used to detect the serum immunoglobulins in weaned piglets.Paraffin section and immunohistochemical method were used to detect the expression and distribution of tight junction protein ZO-1 in intestinal epithelium of piglets,and real-time RCR method was used to detect the expression of tight junction protein ZO-1 mRNA in intestinal epithelium of weaned piglet.The purpose of this Experiment was to investigate the effect of7 S and 11 S on serum IgG subtypes ?IgE levels and intestinal mucosa ZO-1 expression in piglets.Seventy “Duroc×Landrace×Yorkshire” weaned piglets(21 d of age)were randomly divided into seven groups,ten piglets each group.The piglets in control group(Group A)were fed with basic diet,and piglets in groups B,C,D were fed with 500,2000,5000 ?g/kg?w?-conglycinin(7S),respectively,piglets in groups E,F,G were fed with were fed with 500,2000,5000?g/kg?w glycinin(11S)at 24 d,respectively.The blood samples were collected at the ages of 24,26,28 and 30 d,and then the immunoglobulin levels were measured.At the end of this test,five piglets in each group were bled to death and collected each segment of the small intestine in which front,middle and posterior segments of jejunum were selected.All the samples were placed in 4% paraformaldehyde and stored in liquid nitrogen which was used to detect the expression and distribution of the intestinal epithelial tight junction protein ZO-1 and the relative expression of each small intestine ZO-1 mRNA.The results were as follows:1.The serum levels of IgG,IgG1,IgG2,IgG3,IgG4 and IgE in the piglets sensitized by soybean protein antigen were increased,and groups B,C,D were higher than groups E,F,G.2.The relative expression levels of ZO-1 in intestinal epithelium of piglets sensitized by soybean protein antigen were reduced,and it was negatively correlated between the addition amount of 7S,11 S and the relative expression levels of ZO-1.The relative expression levels of ZO-1 in groups B,C,D were less than in groups E,F,G.3.The relative expression levels of ZO-1 mRNA were decreased in each test group,and it was negatively correlated between the addition amount of 7S,11 S and the relative expressionlevels of ZO-1 mRNA.The relative expression of ZO-1 mRNA in groups B,C,D was less than in groups E,F,G.The results showed that ?-conglycinin and glycinin had significant sensitization on weaned piglets,the levels of serum IgG,IgG1,IgG2,IgG3,IgG4 and IgE in the piglets sensitized by ?-conglycinin and glycinin were increased,and the relative expression levels of ZO-1 was reduced in the intestinal epithelial.
