Due to the wide variety of local ducks in our country,the research on breeding and utilization of local duck has became hot.It is very meaningful to study genetic uniformity of feather color on that feather color is not only the important quality traits of different ducks but also the feature of different economic values,thus when we breeding ducks we concentrate more on the color regularity degree of feather which is decided by melanin content.In this experiment,through the establishment of the Chaohu duck basic group and continuous selected for 2 generations,the specialized line of Chaohu Duck was built.Based on this,we started the research on the hybridization generation of Chaohu duck and Cherry Valley duck including separation of feather color and related gene expression and analyzed the principle of inheritance and formation mechanism of feather color.The results were illustrated as follows:1 The group of Chaohu duck contained both line L(28 families)and line H(50families).Breeding line L focused mainly on the early growth combined with plumage while line H was based on the adult weight and laying rate of 40 wk of age.After continuous breeding and selection for 2 generations,compared with the 0generation,the weight of 12 wk of age ducks increased by 12% which is 2235 g and male and female born weight of line L increased by 8.8% and 7.1% respectively which were 50.8g and 51.2g respectively.The 40 wk of age ducks' egg production reached 112 pieces increased by 36.6% and fertilization rate of line H reached 95%increased by 7.9%.The work of breeding Chaohu duck achieved initial success.2 The plumage phenotypes of the Chaohu duck and Cherry Valley duck hybrid F1 generation and F2 generation in front of the 2-week-old showed black feather and gray and yellow.However,the sixth week for youth feather replacement,only black white feather and feather phenotype was observed.Breeding plumage color separation to the sixth week of statistics: in F1 group with ratio of 9:6(?2= 0.28,p>0.05)and F2 groups with ratio of 9:7(?2 = 0.01,p>0.05).The black-white plumage appeared at the neck and wings.3 The expression of TYR gene in hemp feather duck of F1 and F2 reached the peak which is 4.01(p<0.05)and 5.43(p<0.01)in the fourth week.Expression trends of the duck group both hemp plumage and black-white plumage were consistent during each week.The expression of MITF gene in hemp feather of F1 generationreached 0.83(p<0.05)and 1.05(p<0.01)in fourth and fifth weeks significantly higher than the Cherry Valley Duck and Chaohu duck at same time.The expression of black-white feather duck was consistent with age change.The expression of KIT gene in hemp feather and black-white duck were 2.44(p<0.01),1.9(p<0.05)KIT,which were significantly higher than the Cherry Valley Duck and Chaohu duck at same time.The trend of expression contains TYR gene,MITF gene and KIT gene in hemp plumage and black-white plumage belonged to F1 and F2 seemed consistently.Moreover,all the peaks of expression appeared in the 4 or 5 wk of age.
